摘  要:目的探究铜绿假单胞菌构建新西兰兔脓胸模型的条件,为进一步研究铜绿假单胞菌在胸腔环境中的生长特性、耐药机制及其相应的诊疗策略提供基础.方法将24只新西兰兔随机分成实验组和对照组,对照组8只,实验组16只;通过胸腔穿刺置管术于新西兰兔右侧第六肋间隙将引流管置入胸腔,实验组向胸腔内注入2ml/kg的10^5~10^6CFU/ml铜绿假单胞菌标准菌株PAO1菌液;对照组向胸腔内注入等量LB肉汤;将引流管埋入胸腔内,在清洁级环境下正常饲养.于术后第4天解剖新西兰兔,取胸膜组织进行HE染色,观察胸膜增厚及炎症细胞浸润情况,并取胸腔积液/脓絮状物进行革兰染色观察铜绿假单胞菌;取引流管进行细菌培养及活菌计数以证明铜绿假单胞菌感染所致的脓胸模型是否建立成功.结果共有22只新西兰兔完成实验,实验组新西兰兔患侧胸腔中形成不等量的胸腔积液/脓絮状物,广泛存在组织粘连.胸膜组织HE染色显示实验组的新西兰兔胸膜明显增厚,并伴随大量炎症细胞浸润.实验组新西兰兔的胸腔积液/脓絮状物革兰染色可见革兰阴性杆菌.实验组留置的引流管中可培养出大量铜绿假单胞菌.结论用LB肉汤作为铜绿假单胞菌悬液的溶剂,将细菌注入胸腔可成功建立新西兰兔铜绿假单胞菌脓胸模型.Objective To explore the conditions in the construction of experimental empyema by Pseudomonas aeruginosa in New Zealand rabbits,so as to provide a basis for further study on the growth characteristics,drug resistance mechanism and corresponding diagnosis and treatment strategies of Pseudomonas aeruginosa in the thoracic environment.Methods Twenty-four New Zealand rabbits were randomly divided into the experimental group(n=16)and the control group(n=8).The drainage tube was placed into the thoracic cavity in the sixth intercostal space on the right side of the New Zealand rabbits by thoracentesis.Injected 2ml/kg of 10^5~10^6 CFU/ml Pseudomonas aeruginosa standard strain PAO1 into the thoracic cavity of the experimental group;and injected the same dose of LB broth into the thoracic cavity of the control group.The drainage tube was embedded into the thoracic cavity.The animals were raised normally in clean environment.On the fourth day after operation,rabbits were dissected,and pleural tissues were taken for HE staining to observe pleural thickening and inflammatory cell infiltration.Pleural effusion/pussy was taken for Gram staining to observe Pseudomonas aeruginosa.Tube culture and viable counts were performed to demonstrate the success of the establishment of an experimental empyema caused by Pseudomonas aeruginosa infection.Results A total of 22 New Zealand rabbits completed the experiment.The experimental group developed unequal pleural effusion/pus in the thoracic cavity of the chest.The HE staining of the tissue adhesion pleural tissue showed that the New Zealand rabbits pleura increased significantly in the experimental group and accompanied by a large infiltration of inflammatory cells.Gram-negative bacilli were observed in the pleural effusion/pus gram stain of New Zealand rabbits in the experimental group.A large amount of Pseudomonas aeruginosa was cultured in the drainage tube left in the experimental group.Conclusion The experimental empyema by Pseudomonas aeruginosa in New Zeland rabbits can be succe
