摘 要:目的:建立高效液相色谱(HPLC)法检测人血浆中替考拉宁浓度的方法。方法:采用Waters System-C18色谱柱,以乙腈:0.01mol/L磷酸二氢钠缓冲液(25:75,V/V,H3PO4调pH值为2.53)为流动相,柱温40 ℃,流速1.2 mL/min;进样量20 L;检测波长215nm。结果:本方法得到标准曲线方程为y = 0.053x - 0.021(R2= 0.998),人血浆中替考拉宁浓度在3.125~100 mg/L范围内具有良好的线性关系。回收率大于92%,精密度和稳定性的RSD均小于8%。结论:本研究建立的替考拉宁血药浓度HPLC测定法专属性好、操作简单、准确度高、重现性好、回收率和灵敏度高,适用于生物样本分析研究,可满足替考拉宁药动学研究的需要。Objective: To establish a HPLC method to determine teicoplanin in human plasma. Methods:Chromatographic conditions included Waters C18 column, acetonitrile-0.01 mol/L NaH2PO4(25:75,V/V,pH adjusted to 2.53 with H3PO4)as mobile phase, 40 ℃ column temperature, 215 nm detective wavelength, and 1.2 mL/min flow rate. Results:This method generated a standard curve of teicoplanin with good linearty in the concentration of 3.125~100 mg/L(R2= 0.998),the recovery was over 92%, and the precision and the stability were also qualified as their RSD were below 8%. Conclusion:We believe this method is sensitive, simple, accurate, precisely, and stable. It could be used for the determination of teicoplanin in plasma and fulfill the needs of clinical pharmacokinetics for teicoplanin.
