摘　　要：目的探讨RASGRP1 rs7403531基因多态性对瑞格列奈治疗2型糖尿病(T2DM)患者疗效的影响。方法用直接测序法对78名T2DM患者进行RASGRP1 rs7403531基因型分析。初诊的T2DM患者均给予瑞格列奈每次1mg,tid,连续口服8周。用药前和用药第8周末收集受试者静脉血样,测定相关糖脂代谢等实验室指标。结果治疗8周后,CT+TT基因型组餐后2 h血糖(2-h PG)为(11. 86±3. 17) mmol·L^-1,明显高于CC基因型组的(10. 32±2. 41) mmol·L^-1,差异有统计学意义(P <0. 05),且CT+TT基因型组2-h PG值降低幅度为(4. 42±3. 95) mmol·L^-1,显著低于CC基因型组(7. 07±3. 64) mmol·L^-1,差异有统计学意义(P <0. 01)。结论 RASGRP1rs7403531基因多态性与瑞格列奈疗效相关,CC基因型患者疗效更好。Objective To investigate the impact of RASGRP1 rs7403531 polymorphism on repaglinide efficacy in the patients with type 2 diabetes mellitus( T2DM). Methods A total of 78 patients with T2DM were included to determine their genotypes for the RASGRP1 rs7403531 polymorphism by direct DNA sequencing. Newly-diagnosed T2DM patients were orally administrated 1 mg repaglinide three times per day for 8 consecutive weeks. Blood was collected to determine metabolism-related indicators before and after treatment. Results After treatment,the levels of postprandial plasma glucose( 2-h PG) were higher in T2DM patients with CT + TT genotypes( 11. 86 ± 3. 17) mmol·L^-1,compared with CC genotypes( 10. 32 ± 2. 41) mmol·L^-1,( P < 0. 05). Additionally, T2DM patients with RASGRP1 rs7403531 CC genotypes( 7. 07 ± 3. 64) mmol·L^-1 showed more decrease in terms of 2-h PG compared with those with CT + TT genotypes( 4. 42 ± 3. 95) mmol·L^-1,( P < 0. 01). Conclusion RASGRP1 rs7403531 genetic polymorphism may have an effect on repaglinide therapeutic efficacy,and T2DM patients with RASGRP1 rs7403531 CC genotypes showed better efficacy.
