摘 要:目的建立一种同时测定金卷升板胶囊中没食子酸、绿原酸、黄芩苷、芦荟大黄素和大黄素甲醚含量的高效液相方法。方法色谱柱:Diamonsil C18柱(250. 0 mm×4. 6 mm,5μm),流动相:0. 1%乙酸水-甲醇,梯度洗脱,检测波长:275 nm,流速:1 mL·min^-1。考察该方法的专属性、标准曲线与定量下限、精度度与回收率、稳定性。结果没食子酸、绿原酸、黄芩苷、芦荟大黄素和大黄素甲醚分别在1. 07~17. 21,4. 92~79. 34,5. 08~83. 69,1. 34~23. 14和2. 58~42. 07μg·mL^-1内线性关系良好(均r≥0. 999 1)。专属性和精密度良好,样品24 h内稳定性良好。高、中、低浓度的平均回收率范围分别是94. 41%~99. 36%,96. 51%~101. 57%和94. 36%~102. 06%(RSD≤2. 17,n=9)。结论该方法稳定、快速,可为金卷升板胶囊的质量控制提供依据。Objective To develop a HPLC method for determination of the concentrations of five components in Jin - Juan - Sheng - Ban capsules. Methods Chromatographic separation was carried out on Diamonsil C18 ( 250. 0 mm × 4. 6 mm,5 μm) column. The mobile phase was 0. 1% acetic acid in water ( A)- methanol ( B) with gradient elution. The detection wavelength was 275 nm, and the flow rate was 1 mL·min^-1 . The specificity,standard curve,accuracy,sample recovery, and stability of the method were investigated. Results The calibration curves of gallic acid,chlorogenic acid,baicalin,aloe emodinn and physcion were linear in the ranges of 1. 07 - 17. 21,4. 92 - 79. 34, 5. 08 - 83. 69,1. 34 - 23. 14,2. 58 - 42. 07 μg·mL^-1 ( r≥0. 999 1). Specificity and precision were good,and the sample was stable in 24 h. The average recoveries of high,medium and low concentrations were 94. 41%- 99. 36%, 96. 51%- 101. 57%, 94. 36%- 102. 06%( RSD≤2. 17,n = 9). Conclusion The established method was stable and fast,which can be used for the quality concrol of Jin - Juan - Sheng - Ban capsules.
