摘 要:【目的】从显色方法、提取条件、测定方法 3方面研究确立乌灵胶囊总黄酮含量的测定方法。【方法】采用亚硝酸钠(NaNO2)-硝酸铝[Al(NO3)3]-氢氧化钠(NaOH)显色法、三氯化铝(AlCl3)显色法、盐酸-镁粉显色法3种显色方法,以正交法优化提取条件,紫外分光光度法测定乌灵胶囊总黄酮含量。【结果】最优提取条件:在85℃水浴条件下,用体积分数90%乙醇100 mL水浴回流提取2 h。含量检测以芦丁为对照品,采用以NaNO2-Al(NO3)3-NaOH显色的可见分光光度法在510 nm处测定乌灵胶囊中总黄酮含量,结果显示,芦丁对照品在5.5~55μg·mL-1范围内线性关系良好,线性回归方程:Y=0.0130X-0.017 6(R2=0.999 7),平均回收率为99.420%(n=6),相对标准偏差(sR)为1.270%。【结论】AlCl3显色法、盐酸-镁粉显色法不能用于乌灵胶囊总黄酮的测定;NaNO2-Al(NO3)3-NaOH显色法准确度、精密度高,重复性好,是乌灵胶囊总黄酮定量分析的合理方法。Objective To establish a determination method for the content of total flavonoids in Wuling Capsules in the 3 aspects of colorimetric method,extraction condition and determination method. Methods Three colorimetry methods including NaNO2-Al(NO3)3-NaOH colorimetric method, AlCl3 colorimetric method, and HCl-Mg colorimetric method were used,the extraction conditions were optimized by orthogonal test,and the content of total flavonoids in Wuling Capsules was determined by ultraviolet spectrophotometry. Results The optimal extraction conditions for extraction were as follows:under water bath condition at 85 ℃,reflux extraction was carried out for 2 h with 100 mL of volume fraction of 90% of ethanol. With rutin as reference substance, the content of total flavonoids in Wuling Capsules was determined by NaNO2-Al(NO3)3-NaOH coloration visible spectrophotometry at 510 nm. The results showed that rutin had a good linear relationship in the range of 5.5 ~55 μg·mL-1,the regressive equation was Y = 0.013 0 X-0.017 6(R2= 0.999 7), the average recovery was 99.420%(n = 6),and sR was 1.270%. Conclusion AlCl3 chromogenic method and HCl-Mg chromogenic method are unsuitable for the determination of total flavonoids in Wuling Capsules. NaNO2-Al(NO3)3-NaOH colorimetry method has high accuracy and precision, good repeatability, and is reasonable for the determination of total flavonoids in Wuling Capsules.
