摘 要:目的探讨Flic蛋白作为免疫调节剂对低剪切力所致动脉粥样硬化的影响及机制。方法雄性新西兰大耳白兔18只随机分为3组:对照组(Con组,n=6)、低剪切力组(LSS组,n=6)、低剪切力+Flic蛋白组(Flic组,n=6),通过颈动脉套管法建立低剪切力致动脉粥样硬化模型。应用彩色多普勒超声检测颈动脉血管狭窄程度及阻力指数RI值,HE染色评价斑块面积,ELISA方法检测血浆IFN-γ及IL-4水平。结果与Con组比较,Flic组和LSS组动脉血管狭窄程度、阻力指数RI值及斑块面积均明显增加(P<0.05);与LSS组比较,Flic组动脉血管狭窄程度及阻力指数RI值明显降低(P<0.05),斑块面积明显缩小(P<0.05)。与Con组比较,Flic组和LSS组血清IFN-γ水平明显升高(P<0.05),IL-4水平明显降低(P<0.05),IFN-γ/IL-4比值升高(P<0.05);与LSS组比较,Flic组IFN-γ水平明显降低(P<0.05),IL-4水平明显升高(P<0.05),IFN-γ/IL-4比值明显降低(P<0.05)。结论Flic蛋白能够减轻低剪切力所致动脉粥样硬化,其机制可能为调节Th1/Th2平衡。Objective To investigate if the Flic protein have protective effect and its possible immune mechanism in atherosclerosis caused by low shear force.Methods Totally 18 male New Zealand big ear rabbits were selected,and randomly divided into control group (Con group),low shear stress group (LSS group),and low shear stress+Flic protein group (Flic group),6 rats in each group.We establised animal model of low shear force through the method of carotid artery casing.When the model was established successfully,all the rabbits were treated with high-fat diet 12 weeks in order to accelerate the formation of atherosclerosis in low shear zone.Ultrasound of carotid artery were performed on rabbits with ultrasound imaging system,degree,color Doppler flow imaging and vascular imaging to detect the stenosis of carotid artery.The resistance index RI,HE was used to measure plaque size.While enzyme-linked immunosorbent assay method was used to measure serum Th1 cells representative factor interferon-γ(INF-γ) and Th2 cells representative factor interleukin-4 (IL-4).Results Compared with Con group,the arterial stenosis rate,RI values and plaque size were significantly increased in the LSS group and Flic group,a statistically difference ( P <0.05).Compared with the LSS group,the arterial stenosis rate,RI values and plaque size significantly smaller in the Flic group,statistically significant difference ( P <0.05).Compared with Con group,the levels of serum Th1 cells representative factor INF-γ increased significantly,while the levels of serum Th2 cells representative factor IL-4 decreased significantly in the LSS group and Flic group.The ratio of INF-γ/IL-4 increased,statistically significant difference ( P <0.05).Compared with LSS group,the levels of serum Th1 cells representative factor INF-γ significantly decreased,while the levels of serum Th2 cells representative factor IL-4 significantly increased in the Flic group.The ratio of INF-γ/IL-4 decrease obviously;increased statistically significant difference ( P <0.05).Conclusi
