摘 要:目的 观察有氧运动对2型糖尿病大鼠骨表型的影响。方法 以50只6周龄雄性SD大鼠为研究对象,通过给大鼠喂饲高脂高糖饲料、注射链脲佐菌素建立糖尿病模型,经过跑台运动后分为正常对照(control,C)组、糖尿病对照(diabetes mellitus,DM)组和糖尿病运动(diabetes mellitus with exercise,EDM)组。后micro-CT检测大鼠股骨骨密度和骨微含量,real-time PCR定量检测大鼠胫骨成骨、成脂、破骨相关基因的表达量。结果 建模成功后DM组、EDM组大鼠的体质量显著低于C组( P <0.05),第1周到第4周DM组和EDM组大鼠的体质量较之于C组均显著偏低( P <0.01),第3周EDM组大鼠体质量低于DM组( P <0.05)。C组、DM组和EDM组大鼠左腿股骨骨密度无差异;EDM组大鼠股骨骨小梁相对体积(trabecular volume,BV/TV)、骨小梁厚度(trabecular thickness,Tb.Th)、骨小梁数量(trabecular number,Tb.N)都显著低于C组( P <0.05)。DM组Runx2 mRNA表达低于C组( P <0.05);EDM组碱性磷酸酶(alkaline Phosphatase,ALP)mRNA表达高于C组和DM组( P <0.05),Osterix mRNA表达高于C组( P <0.05)且显著高于DM组( P <0.01),RANKL mRNA表达低于C组( P <0.05)。各组大鼠胫骨中CEBP、Ctsk、OPG、RANK mRNA表达均未出现差异。结论 4周的跑台运动可以促进2型糖尿病大鼠骨形成、有效地抑制骨吸收和脂肪形成,但并没有明显反映在骨量上。Objective To explore the change of bone phenotypes of type 2 diabetes mellitus rats. Methods We chose 50 6-week old male SD rats, and established diabetes mellitus model rats by high-fat diet and injecting streptozocin. Then the rats were randomly divided into control (C) group, diabetes mellitus (DM) group and diabetic mellitus exercise (EDM) group after treadmill exercise. Bone mineral density (BMD) and bone microarchitecture parameters of rats' femur were measured by a micro-CT scanner and osteoblast, adipocyte, and osteoclast-selective genes were measured by quantitative testing. Results After modeling, body mass of rats in DM and EDM group were significantly lower than C group ( P <0.05). From first week to the fourth week, body mass of rats in DM and EDM group was significantly lower than C group ( P <0.01);the third week body mass of rats in EDM group was lower than the DM group ( P <0.05). BMD at left femurs had no significant difference among these groups. The value of trabecular volume (BV/TV), trabecular thickness (Tb.Th), and trabecular number (Tb.N) in EDM group were lower than C group ( P <0.05). Runx2 mRNA expression in DM group was lower than C group ( P <0.05). Alkaline phosphatase (ALP) mRNA expression in EDM group was higher than C and DM group ( P <0.05). Osterix mRNA expression in EDM group was higher than C group ( P <0.05) and DM group ( P <0.01). Compared with C group, RANKL mRNA expression was lower in EDM group ( P <0.05). CEBP,Ctsk,OPG,RANK mRNA of rats' left femurs had no significant difference in each group. Conclusion Four weeks aerobic exercise can improve osteogenesis and inhibit bone resorption and adipogenesis of type 2 diabetes mellitus rats, but this effect had no obvious effects on bone mass.
