摘 要:目的 探讨白藜芦醇对脑出血和小胶质细胞的保护作用。方法 采用Rosenberg法构建大鼠脑出血模型,并给予白藜芦醇(30mg/kg),分组为对照组、脑出血模型组和脑出血模型+白藜芦醇组,并检测三组大鼠神经功能评分、脑组织含水量、脑组织血肿体积;培养SD乳鼠小胶质细胞,给予25μM的白藜芦醇处理,采用CCK-8实验和ELISA试剂盒检测两组细胞活性和炎性因子水平。结果与对照组相比,脑出血模型组大鼠神经功能评分升高、脑组织含水量增加、脑组织血肿体积增加,而给予白藜芦醇能够逆转这些变化。此外,给予白藜芦醇不能改变小胶质细胞活性,但能降低小胶质细胞炎性因子水平。结论白藜芦醇能够改善脑出血后的炎性损伤,可应用于临床治疗脑出血。Objective To investigate the protective effect of resveratrol on intracerebral hemorrhage and microglia.Methods The rat model of intracerebral hemorrhage was established by Rosenberg method,and resveratrol(30 mg/kg)was administered into the rats.The rats were divided into the control group,intracerebral hemorrhage model group,intracerebral hemorrhage model + resveratrol group.Rat neurological function score,brain tissue water content,brain tissue hematoma volume of SD rats were detected.The microglia were cultured and treated with25 μM resveratrol.CCK-8 assay and ELISA kit were used to detect the cell viability and the level of inflammatory factor respectively.Results Compared with the control group,the neurological function score,brain tissue water content and brain tissue hematoma volume of the intracerebral hemorrhage model group was increased,while resveratrol could reverse these changes.In addition,the resveratrol was not able to change the cell viability of microglia,but could reduce the level of inflammatory factor of microglial.Conclusion Resveratrol can improve the inflammatory injury after intracerebral hemorrhage and may be used for clinical treatment of intracerebral hemorrhage.
