摘 要:对福建省新流行谱系3的PRRSV进行分离,并对其分子生物学特性进行初步研究,为新流行毒株的防治提供依据和参考。分离的毒株通过RT-PCR和IFA对PRRSV分离毒株进行鉴定,并对其ORF5基因进行序列测定分析。结果显示,分离株能够在Marc-145细胞上增殖并产生典型的细胞病变,被抗PRRSV的特异性单克隆抗体所识别,具有特异性免疫荧光,其TCID50为105.3,将其命名为FJFQ1805。分离株ORF5基因与北美型PRRSV参考株核苷酸同源性为83.9%~92.2%,与谱系3病毒株QYYZ的同源性最高,为92.2%。系统进化树分析表明,该分离株属于北美型,与QYYZ、GM2和FJFS等谱系3的病毒株属于同一分支。综上,所分离的FJFQ1805毒株为谱系3的病毒株。PRRSV strain of the new epidemic lineage 3 in Fujian Province was isolated using Marc-145 cells and its biological characteristics were preliminarily studied, providing the basis and reference for the prevention and control of new epidemic strains. The virus isolate was determined by RT-PCR and confirmed by immunofluorescence assay (IFA), and the ORF5 gene was sequenced and analyzed. The results showed that PRRSV typical CPE was observed on Marc-145 cells and it was detected by anti-PRRSV spe- cific monoclonal antibodies by IFA,The TCID 50 of cultured virus was 10 5. 3 . The PRRSV strain was named as FJFQ1805. Sequence a- nalysis of ORF5 showed that the isolate shared 83. 9%~92. 2% homology with reference isolates,and a high identity of 99. 2% with QYYZ. The phylogenetic tree analysis showed that the isolate belonged to NA and is closely related to FJFS, QYYZ and GM2. In conclusion, FJFQ1805 belongs to lineage 3 PRRSV.
