摘 要:为研究猪繁殖与呼吸综合征病毒(PRRSV)在广东流行毒株的分子遗传变异情况和发展趋势,通过RT-PCR方法,对2016-2017年采自广东省各地906份疑似病料进行PRRSV检测,并对其中9株PRRSV的NSP2和ORF5基因进行测序及遗传变异分析。结果显示,有399份样品被检出为阳性,阳性率达到44%;通过NSP2和ORF5基因序列比对和遗传进化分析,发现广东流行毒株主要为美洲型PRRSV,且全部9株与高致病性猪繁殖与呼吸综合征病毒(HP-PRRSV)高度同源;其中7株的NSP2蛋白存在与HP-PRRSV相同的1+29个氨基酸的不连续缺失特征;9株GP5蛋白的2个重要抗原相关区域的氨基酸位变化特征与HP-PRRSV一致,与美洲型代表株VR-2332相比变异明显。结果表明,PRRSV在广东流行的情况仍比较严重,当前HP-PRRSV依然是广东的优势病毒株,提示对PRRSV的流行和变异进行持续监测是有必要的,还应加强对其致病机理和新型疫苗研发等的深入研究。To investigate the molecular genetic variations and development trend of Porcine Reproductive and Respiratory Syn- drome Virus (PRRSV) in Guangdong province, 906 suspected clinical samples to be infected with PRRSV from pig farms in Guang- dong province during 2016 to 2017 were collected for RT-PCR detection. The NSP2 and ORF5 genes of 9 PRRSV strains from Guang- dong, respectively, were sequenced and analyzed. The results showed that 399 samples were positive for PRRSVs, with a positive rate of 44%. The phylogenetic analysis revealed that most of the prevalent strains were the American genotype. All of the 9 strains were highly homologous with HP-PRRSV. Seven of 9 strains were the same feature with HP-PRRSV in NSP2 proteins characters by a discontinuous deletion of 1 +29 amino acids. Moreover, no similar to the NADC30 strain was found, which contained a discontinuous deletion of 131 amino acids with HP-PRRSVs in NSP2 proteins. All of 9 strains were the same amino acids variation characteristics with HP-PRRSV in 2 important antigen-related regions of GP5 proteins, which were significant variations compared with the American representative strain VR-2332. In conclusion, the epidemic of PRRSVs remained serious and HP-PRRSVs were still the dominant strains in Guangdong, suggesting that more attention should be continuously paid to monitor the pathogenic epidemiology and genetic variation of PRRSVs. Furthermore, additional research should be conducted on pathogenesis and the development of new vaccines of PRRSVs.
