摘  要:利用生物碱盐酸盐易溶于水这一特性,用稀盐酸溶液进行红茂草总生物碱的提取试验。先分别进行提取时间、料液比、pH值和温度对红茂草总生物碱提取率影响的单因素试验,在此基础上再从每个单因素中选出3个较好水平进行正交试验,从而得出酸水提取红茂草总生物碱的最佳条件。按照正交试验所确定的最佳提取条件进行总生物碱的提取。以红茂草中主要生物碱异紫堇碱为指标,对提取物进行HPLC检测。结果表明,在温度90℃,提取时间5h,料液比1∶40,pH值1.5为提取红茂草总生物碱的最佳条件。HPLC检测结果显示,提取物中异紫堇碱含量为9.28%。分别占分离菌株的25.6%、27.9%。本试验为该区猪源肺炎克雷伯杆菌的防治提供理论基础。The experiment was conducted on the extraction of total alkaloid of Dicranostigma leptotodum(Maxim) by diluted hy- drochloric acid solution. Firstly, four single factor experiments were conducted respectively to determine the effect of time, tempera- ture, pH value and the ratio of material to liquid on the extraction of total alkaloid of Dicranostigma leptodum(Maxim.) Fedde. On this basis, the optimum conditions for extracting were investigated by using the orthogonal design. The extract was dissolved by meth- anol for HPLC determination using isocorydine as index. The results indicated that the optimum conditions for the extraction of total alkaloid of Dicranostigma leptodum(Maxim.) Fedde were as following: temperature 90 ℃, reaction time 5h, pH 1. 5, and the ratio of material to liquid was 1∶ 40. The HPLC analysis showed that the content of isocorydine was 9. 28%.
