摘  要:采用农杆菌介导法将黄瓜表皮毛基因Tril转入黄瓜‘新泰密刺’品种,并研究了6-苄氨基腺嘌呤(6-BA)和卡那霉素(Kan)分别对再生和转化苗筛选效果的影响。结果表明,在黄瓜‘新泰密刺’转化中,Kan抗性苗出芽率为9.67%,生根率为3.67%,转化株阳性率为0.33%;分化培养基中6-BA适宜浓度为0.5mg/L,出芽率最高(76.33%),平均出芽天数最短(19.66d);生根培养基中Kan浓度为60mg/L时筛选效果最好。3种筛选方式的研究表明,仅在生根培养基中添加Kan(60mg/L)筛选,其转化周期最短(45d),阳性率最高(达4.0%)。In this study,the cucumber Tril gene was transferred into the genome of‘Xintai Mici’,a cucumber cultivar,mediated by Agrobacterium.In addition,the effects of 6-benzylaminopurine(6-BA) and kanamycin(Kan) on the regeneration and transformant screening were studied.The results showed that the regeneration rate of Kan resistant shoots was 9.67%,the rooting rate was 3.67% and the positive rate was 0.33% in the transformation of ‘Xintai Mici’.In the differentiation medium,the optimal concentration of 6-BA was 0.5 mg/L,which produced the highest rate of shoot regeneration(76.33%) and the average shooting days(19.66 d).In the rooting medium,the optimal concentration of Kan for transformant screening was 60 mg/L.The analysis of three screening methods showed that the transformation period was the shortest(45 d) and the positive rate was the highest(4.0%) when the rooting medium contained 60 mg/L Kan.
