摘  要:目的 探究睡眠剥夺过程中小鼠海马组织内脑源性神经营养因子(brain derived neurotrophic factor,BDNF)、白细胞介素4(Interleukin-4,IL-4)的动态变化及相关性,从而为睡眠剥夺对海马学习记忆功能的影响提供可能的理论依据。方法 (1)6~8周龄C57BL/6J雄性小鼠随机分为正常对照组(Cage control,CC)、快速眼球运动(REM)睡眠剥夺1 d组(REM sleep deprivation 1 day,SD 1 d)、REM 睡眠剥夺2 d组(SD 2 d)、REM 睡眠剥夺3 d组(SD 3 d),每组12只小鼠。CC组同笼饲养1 w后于9:00AM灌注处死,SD 1 d组、SD 2 d组、SD 3 d组经睡眠剥夺1 d、2 d和3 d后均于9:00AM灌注处死。(2)采用ELISA方法观察各组小鼠海马组织中BDNF、IL-4的表达水平。结果 (1)ELISA显示SD 1 d组小鼠海马中BDNF、IL-4较CC组表达水平升高,差异具有统计学意义( P 值分别为0.029、<0.001 );SD 2 d组BDNF、IL-4较CC组表达无明显差异( P 值分别为>0.5、0.13);SD 3 d组BDNF、IL-4较CC组表达水平明显降低,差异具有统计学意义( P 值均为<0.001)。(2)Pearson相关性分析显示睡眠剥夺过程中小鼠海马中IL-4、BDNF蛋白表达变化呈正相关性, P <0.001,相关系数 r =0.863。结论 睡眠剥夺后BDNF、IL-4表达量的变化可能是睡眠剥夺后造成神经损伤的可能机制之一。Objective To investigate the changes of brain-derived neurotrophic factor (brain derived neurotrophic factor,BDNF) and interleukin-4 (Interleukin-4,IL-4) in the mice hippocampus during sleep deprivation. And a preliminary study was conducted on the possible mechanism of action between the IL-4 and BDNF,which provided a possible theoretical basis for the effects of sleep deprivation on hippocampal learning and memory functions. Methods 1.C57BL/6J male mice were randomly divided into normal control group (Cage control,CC),REM sleep deprivation 1 d group (REM sleep deprivation 1 day,SD 1 d),REM sleep deprivation 2 d group (SD 2 d),REM sleep deprivation 3 d group (SD 3 d),12 mice in each group. After 1 week of cage feeding,CC group was killed at 9:00AM. SD 1 d group SD 2 d group and SD 3 d group was killed after 1、2 and 3 days of sleep deprivation at the same time respective. 2.The protein expression of BDNF,IL-4 in the hippocampus of each group were observed by ELISA. Results 1.ELISA demonstrated that the expression of BDNF,IL-4 in the hippocampus of SD 1 d group was higher than those in CC group. The difference was statistically significant ( P =0.029, P <0.001,respectively);Compared with the CC group,BDNF,IL-4 in SD 2 d group were no significant difference ( P >0.5, P =0.13,respectively);SD 3 d group had significantly lower expression of BDNF,IL-4 than the CC group ( P <0.001). 2.Pearson correlation analysis showed that there was a positive correlation between the changes of IL-4 and BDNF protein expression in hippocampus during sleep deprivation, P <0.001,correlation coefficient r =0.863. Conclusions The changes of BDNF and IL-4 expression after sleep deprivation may be one of the possible mechanisms of nerve damage after sleep deprivation.
