摘  要:目的观察孕酮在大鼠脑损伤中的作用,进一步研究孕酮神经保护作用的分子机制。方法120只大鼠,随机分为假手术组、脑损伤组及孕酮治疗组。按照改良的Feeney自由落体损伤装置制作大鼠脑损伤模型;采用酶联免疫吸附法(ELISA)测定脑组织匀浆中S100B蛋白的含量、神经功能及脑水肿测定。结果脑损伤后脑损伤组24hS100B蛋白表达、mNSS检测评分、脑组织含水量明显高于假手术组(P<0.05),而孕酮治疗组S100B蛋白表达、mNSS检测评分、脑组织含水量明显低于脑损伤组(P<0.05)。结论孕酮可通过降低脑组织S100B蛋白的表达,降低脑水肿,改善脑损伤后神经功能恢复。Objective To observe the effects of progesterone on brain injury in rats,and to investiagte further the molecular mechanism of neuroprotective effects of progesterone.Methods A total of 120 rats were randomly divided into three groups:sham operation group,brain injury group and progesterone treatment group.The rat models with brain injury were established by the modified Feeney free fall injury device.The levels of S100B protein in brain homogenate were detected by enzyme linked immunosorbent assay(ELISA),and the nerve function cerebral edema were observed and compared among the three groups.Results At 24h after brain injury,the expression levels of S100B protein,mNSS test scores and brain tissue water content in brain injury group were significantly higher than those in sham operation group(P<0.05),however,which in progesterone treatment group were significantly lower than those in brain injury group(P<0.05).Conclusion Progesterone can reduce the expression of S100B protein in brain tissue of rats,reduce cerebral edema to improve the recovery of nerve function after brain injury.
