老年宫颈癌患者血清NO和Gal-9检测及与凋亡指数的相关性分析

摘　　要：目的检测老年宫颈癌患者一氧化氮(NO)和半乳凝素-9(Gal-9)并分析其与凋亡指数相关性。方法老年宫颈癌患者80例作为宫颈癌组、老年子宫颈上皮不典型增生患者80例作为不典型增生组和正常健康女性80例作为对照组,3组研究对象均取肘静脉分离血清后采用硝酸还原法测定NO含量,采用酶联免疫吸附法(ELISA法)测定Gal-9含量;采用原位末端标记(TUNEL)法检测宫颈细胞凋亡指数。比较3组研究对象血清NO水平、Gal-9水平和凋亡指数差异,采用Spearman秩相关分析血清NO、Gal-9与凋亡指数相关性。结果宫颈癌组宫颈癌患者血清NO、Gal-9及凋亡指数均显著高于不典型增生组和对照组,差异均有统计学意义(P<0.05)。不典型增生组NO、Gal-9及凋亡指数均显著高于对照组,差异有统计学意义(P<0.05)。Spearman秩相关发现宫颈癌患者血清NO、Gal-9与凋亡指数均呈正相关(r值分别为0.373和0.457,P<0.05)。结论老年宫颈癌患者血清NO和Gal-9呈高表达状态,与宫颈癌细胞凋亡指数正相关,可能在老年宫颈癌细胞凋亡中具有一定作用。Objective To detect the expressions of serum nitric oxide(NO)and galectin-9(Gal-9)in elderly patients with cervical cancer,and to analyse the correlation between NO,Gal-9 expressions and apoptotic index.Methods A total of 80 elderly patients with cervical cancer were enrolled as cervical cancer group,and 80 elderly patients with cervical epithelial atypical hyperplasia were enrilled as CIN group,and 80 healthy females were served as control group.The NO levels of all the subjects were measured by nitric acid reduction method,and the Gal-9 content levels were detected by enzyme linked immunosorbent assay(ELISA),and the apoptosis indexes of cervical cells were detected by in situ terminal labeling(TUNEL).Moreover the differences of serum NO levels,serum Gal-9 levels and apoptotic index among the three groups were observed and compared.The correlation between the serum levels of NO,Gal-9 and apoptotic indexes was analyzed by Spearman rank correlation.Results The serum levels of NO,Gal-9 and apoptotic indexes in cervical cancer group were significantly higher than those in CIN group and control group(P<0.05).The levels of NO,Gal-9 and apoptotic indexes in CIN group were significantly higher than those in control group(P<0.05).The Spearman rank correlation showed that there was significantly positive correlation between serum NO,serum Gal-9 and the apoptotic index in patients with cervical cancer(r=0.373,0.457,respectively,P<0.05).Conclusion The NO and Gal-9 are highly expressed in elderly patients with cervical cancer,which is positively correlated with apoptotic index.The NO and Gal-9 may play a certain role in cell apoptosis in elderly patients with cervical cancer.
