摘 要:目的:采用HPLC-DAD-Q-Exactive静电场轨道阱高分辨质谱在线分离鉴定栀子中的主要化学成分。方法:Poroshell 120 SB-C18色谱柱(4.6×150mm,2.7μm),以乙腈-0.1%甲酸水溶液为流动相进行梯度洗脱,质谱采用正、负离子监测模式,以及全扫描及自动触发二级质谱扫描功能,对栀子药材的化学成分进行快速分离和鉴定。结果:根据高分辨质谱数据从栀子甲醇提取物中鉴定了13个主要色谱峰,包括环烯醚萜类8个,二萜色素类5个。结论:建立的HPLC-DAD-Q-Exactive高分辨液质联用技术,为栀子化学成分鉴定提供了一种快速、高效的分析方法。Objective:The main chemical constituents from the fruits of Gardenia jasminoides were identified by HPLC–DAD-Q-Exactive-Orbitrap HRMS.Methods:A Poroshell 120 SB-C18 column(150 mm×4.6 mm,2.7μm)was used.The mobile phase was acetonitrile and water containing 0.1%formic acid by gradient elution.The main peaks in Gardenia jasminoides were rapidly identified by HRMS/MS in the positive and negative ion mode using both full scan and two stage threshold-triggered mass modes.Results:A total of 13 compounds from the roots of Gardenia jasminoides were identified,including 8 iridoids and 5 diterpenes.Conclusion:A rapid and reliable method using HPLC-DAD-Q-Exactive-Orbitrap HRMS/MS is established and can be used to identify the main compounds in Gardenia jasminoides.
