摘 要:目的探讨西黄丸大鼠含药血清对人乳腺癌细胞增殖的作用特点。方法 SD大鼠48只随机分为空白对照组12只及西黄丸0. 5 h组、西黄丸1 h组、西黄丸2 h组、西黄丸3 h组各9只。西黄丸各时间组给予2. 4 g/(kg·d)西黄丸混悬液灌胃,每日2次,空白对照组大鼠给予等体积蒸馏水,连续给药7次。于末次给药0. 5、1、2、3 h后对相应组采血,制备含药血清。MDA-MB-435和MCF-7细胞分别设10%空白血清组、20%空白血清组、30%空白血清组、10%西黄丸血清组、20%西黄丸血清组和30%西黄丸血清组,每组6个复孔。加入相应含药血清培养后,采用MTT方法检测不同采血时间点、不同浓度的西黄丸大鼠含药血清对乳腺癌细胞株增殖的抑制作用。结果与其他时间点相比,西黄丸大鼠各浓度含药血清作用24、48 h均为采血时间2 h含药血清对MDA-MB-435细胞的抑制增殖作用最强;西黄丸大鼠各浓度含药血清作用24、48、72 h均为采血时间3 h含药血清对MCF-7细胞的抑制增殖作用最强(P <0. 05);且均为30%西黄丸含药血清对MDA-MB-435、MCF-7两种细胞株增殖的抑制作用最强(P <0. 05)。结论西黄丸采血时间2 h含药血清对MDA-MB-435细胞的抑制增殖作用最强,采血时间3 h含药血清对MCF-7细胞的抑增殖作用最强,且以30%含药血清抑制作用最强。Objective To investigate the action characteristics of Xihuang Pills(西黄丸) containing rat serum on proliferation of human breast cancer cell. Methods A total of 48 SD rats were randomly divided into a blank control group of 12 rats,a Xihuang Pills 0. 5 h group,Xihuang Pills 1 h group,Xihuang Pills 2 h group and Xihuang Pills 3 h group,with 9 rats in each group. Xihuang Pills groups were given 2. 4 g/( kg·d) Xihuang Pills suspension,twice a day. The rats in the blank control group were given an equal volume of distilled water for 7 times. Blood samples were collected from the corresponding groups at the last dose of 0. 5,1,2,and 3 hours to prepare drug-containing serum. MDA-MB-435 and MCF-7 cells were respectively set to 10% blank serum group,20% blank serum group,30% blank serum group,10% Xihuang Pills serum group,20% Xihuang Pills serum group and 30% Xihuang Pills serum group,6 duplicate wells per group. After adding the corresponding drug-containing serum,MTT method was used to detect the inhibitory effect of serum containing Xihuang Pills in different blood collection time points and different serum volume fractions on the proliferation of breast cancer cell lines. Results Compared with other time points,for the concentration of drug-containing serum in Xihuang Pills 24 h and 48 h after administration,the drug-containing serum had the strongest inhibitory effect on MDA-MB-435 cells at 2 h after administration. The drug-containing serum had the strongest inhibitory effects on MCF-7 cells at 3 h after 24,48 and 72 h after administration( P < 0. 05);and both were 30% Xihuang Pills containing serum had the strongest effects on the proliferation of MDA-MB-435 and MCF-7 cell lines( P < 0. 05). Conclusion The 2 h drug-containing serum of Xihuang Pills has the strongest inhibitory effects on MDA-MB-435 cells. The drug-containing serum for 3 h has the strongest inhibitory effect on MCF-7 cells and 30% drug-containing serum had the most strongest inhibitory effects.
