摘 要:目的研究京尼平苷对db/db小鼠血糖和体重的影响。方法选取8周龄db/db鼠为模型鼠,以同周龄C57BL/6N鼠为对照组;将模型小鼠随机分为2组:模型组和实验组,每组10只。实验组腹腔注射京尼平苷200 mg·kg^-1·d^-1;对照组和模型组腹腔注射等体积0. 9%NaCl,连续2周。监测小鼠血糖,以酶联免疫吸附法法检测小鼠胰岛素;计算体重增长率;以PET-CT检测小鼠体脂。结果干预2周,对照组、模型组和实验组的血糖分别为(4. 32±0. 14),(21. 95±1. 69)和(16. 87±2. 13) mmol·L^-1;对照组、模型组和实验组的胰岛素分别为(0. 13±0. 04),(3. 47±0. 47)和(3. 16±0. 33)μg·L^-1;对照组、模型组和实验组的体重分别为(25. 91±0. 36),(48. 06±1. 01)和(45. 18±0. 74) g。模型组与对照组比较,上述3个指标的差异均有统计学意义(均P <0. 01);实验组与模型组比较,差异均有统计学意义(均P <0. 05)。结论京尼平苷可降低db/db鼠的血糖和体重,其作用分别与增加胰岛素敏感性和减少体脂有关。Objective To investigate the effects of geniposide on blood glucose and body weight in db/db mice. Methods Eight-week-old db/db mice were selected as model mice,and C57BL/6N mice of the same age were used as control group. The model mice were divided into two groups: model group and experimental group,10 mice in each grop. The experimental group was given intraperitoneal injection of geniposide 200 mg·kg^-1·d^-1,the control group and the model group were intraperitoneally injected with equal volume of normal saline for 2 weeks.The blood glucose of the mice was monitored. The plasma insulin was detected by enzyme linked immunosorbent assay method. The body weight growth rate was calculated;and the body fat of the mice was detected by PET-CT. Results After 2 weeks of intervention,the blood glucose of control group,model group and experimental group were( 4. 32 ± 0. 14),( 21. 95 ± 1. 69),and( 16. 87 ± 2. 13) mmol·L^-1,respectively;the insulin levels of the three groups were( 0. 13 ± 0. 04),( 3. 47 ± 0. 47),and( 3. 16 ± 0. 33)μg·L^-1,respectively;the body weight of the three group was( 25. 91 ± 0. 36),( 48. 06 ± 1. 01),and( 45. 18 ± 0. 74) g,respectively. Comparison between model group and control group,the difference of the factors was significant( all P < 0. 01);comparison between experimental group and model group,the difference of the factors was significant( all P < 0. 05). Conclusion Geniposide can decrease blood glucose and body weight in db/db mice,which is related to improve insulin sensitivity and reducing body fat.
