摘　　要：目的探讨影响肝细胞生长因子(hepatocyte growth factor,HGF)对TGF-β1诱导的肌腱成纤维细胞的增殖、凋亡的作用之因素。方法体外实验取大鼠跟腱成纤维细胞,应用TGF-β1(5 ng/mL)诱导肌成纤维细胞纤维化,然后加入HGF干预,测定各组细胞增殖、细胞中RNA和总蛋白的表达、及肌腱成纤维细胞的凋亡。评价黏着斑激酶(FAK)等对HGF的作用的影响。结果 HGF明显抑制了TGF-β1诱导的大鼠肌腱成纤维细胞的过度增殖,且诱导了肌腱成纤维细胞的凋亡。TGF-β1同时增加了α-平滑肌肌动蛋白(α-SMA)和c-MET的基因表达及蛋白水平来诱导成纤维细胞的增殖。结论 HGF通过诱导细胞凋亡来抑制TGF-β1引起的肌腱成纤维细胞增殖。抑制FAK可以阻止HGF诱导的细胞增殖抑制作用。Objective To investigate the effect of HGF affecting the proliferation and apoptosis of TGF-1-induced fibroblast cellular. Methods Tendon fibroblasts of rats were harvested to primary culture in vitro, which were induced fibrosis response by the addition of transforming growth factor β1(TGF-β1)(5 ng/mL). Under conditions with or without the intervention of hepatocyte growth factor(HGF), cell proliferation, RNA expression, total protein expression, and tendon fibroblast apoptosis were measured. In other experiments, the inhibitors of MMPs, ERK1/2, and FAK were also added into the cell cultures before addition of HGF, and the effects were observed. Results HGF inhibited the proliferative effects of TGF-β1 and induced apoptosis in rat tendon fibroblasts. TGF-β1 simultaneously upgraded α-SMA and c-MET expression level in gene and protein level. Conclusion HGF suppress TGF-β1 induced fibroblasts proliferation by cell apoptosis. Inhibition of FAK block HGF-induced growth inhibition, while Inhibition of ERK1/2 could not block HGF-induced growth inhibition.
