微量法筛选黄芪提取液联合左氧氟沙星体外有效抑菌浓度的研究

摘　　要：黄芪提取液联合左氧氟沙星体外实验表现出协同抑菌的功效,因而筛选出有效地抑菌浓度和抑菌谱对于开发新型体表抑菌药显得尤为迫切。本研究应用微量肉汤稀释法结合棋盘法初步筛选对大肠埃希菌、金黄色葡萄球菌、铜绿假单胞菌、化脓性链球菌等8种临床常见标准菌株的有效联合抑菌浓度。结果表明黄芪提取液联合左氧氟沙星在体外对8种临床常见菌株均表现出协同和相加抑菌能力,金黄色葡萄球菌、鲍曼不动杆菌和变形杆菌FIC指数为0.25,其余5种菌株的FIC指数均为0.5。并且成功筛选出0.125+160μg/mL(黄芪提取液+左氧氟沙星)为有效抑菌浓度且具有较广的抑菌谱,具有作为表皮抗感染药物开发基础,但对大肠埃希菌和鲍曼不动杆菌的抑菌效果欠佳。The combination of astragalus extract and levofloxacin in vitro showed synergistic bacteriostatic effect, so it was urgent to screen the effective inhibitory concentration for the development of new epidermal antiinfective drugs. In this study, the effective inhibitory concentrations of 8 common clinical standard strains, such as E.coli, Staphylococcus aureus, Pseudomonas aeruginosa, were preliminarily screened by microliquid culture method. The results showed that astragalus extract combined with levofloxacin had strong synergistic bacteriostatic ability against 8 common clinical strains in vitro.The FIC index of Acinetobacter baumannii, Staphylococcus aureus and Bacillus proteus was 0.25, and the FIC index of the other 5 strains was 0.5. The concentration of 0.008+160μg/mL (Astragalus extract and levofloxacin) was selected as the effective inhibitory concentration and had a wide spectrum of bacteriostatic activity, which was the basis for the development of epidermal anti-infective drugs. But the bacteriostatic effect of Escherichia coli and Acinetobacter baumannii was not good.
