摘  要:目的:选择不同剂量的低强度激光为干预手段,以6周递增负荷跑台训练诱导大鼠运动性免疫抑制(exercise-induced immunosuppression,EIS),通过动态观测多层面的免疫学指标全面评估低强度激光对EIS的调理效果。方法:8周龄雄性SD大鼠104只,随机分为4组:安静对照组(C)、运动训练组(E)、运动和小剂量激光干预组(E+LL)、运动和大剂量激光干预组(E+HL)。每组均再随机分为0周、2周、4周和6周采样时点组。运动各组进行6周递增负荷跑台训练(第1周速度10 m/min;第2周速度20 m/min,第3周起每周速度递增5 m/min,至第6周为40 m/min,30 min/天,6天/周)。激光干预组采用氦氖激光鼻腔外照射(1次/训练日,2 min/次)。照射功率小剂量组1 mw(6.8 J/cm^2),大剂量组2 mw(13.6 J/cm^2)。分别于训练前(0周)及训练的第2、4、6周末,采集各组大鼠的动脉血检测血细胞分类计数以及补体、免疫球蛋白和炎症因子的含量,评价脾脏淋巴细胞增殖能力,并观察胸腺、骨髓和脾脏组织的形态学结构。结果:1)6周递增负荷跑台训练可诱导大鼠免疫机能呈动态性和整体性的负性改变,包括循环血中的免疫细胞数量、免疫球蛋白和补体含量、促炎和抗炎因子平衡,T淋巴细胞增殖能力,以及中枢和外周免疫器官的组织学形态等;2)小剂量低强度激光照射对EIS的发生发展持续产生抵抗作用。循环血中的免疫细胞(T、NE、LY等)数目、细胞因子(TNF-α、IL-10等)以及T淋巴细胞增殖能力等明显改善;骨髓、胸腺的微观结构未见明显异常;3)大剂量低强度激光照射后血中免疫细胞(WBC、NE)数量和细胞因子(ICAM-1、IL-4、IL-10)含量,以及T淋巴细胞增殖能力明显改善。但血清CRP和TNF-α含量等呈负性改变。结论:低强度激光鼻部照射的血液辐照手段可控制和改善递增负荷训练过程中运动性免疫抑制的发生发展,且小剂量照射效果更佳。Purpose:This study aimed to examine the effect of different doses of laser irradiation on the immunosuppression(EIS)induced by six-week incremental exercise.By dynamic observation of the multilevel immunological indicators,effect of low power laser on exercise-induced immunosuppression was assessed comprehensively.Methods:104 male Sprague Dawley rats,aged 8 weeks,were randomly divided into 4 groups:Control(C),Exercise(E),E+low level laser(E+LL,1 mw,6.8 J/cm^2),and E+high level laser(E+HL,2 mw,13.6 J/cm^2).All rats in exercise-related groups performed an incremental treadmill training protocol:6 days/week,30 min/day,and 6 weeks with incremental velocity(10 m/min in 1st week,20 m/min in 2nd week,with 5 m/min/week increment in the following weeks).He-Ne laser was used for laser treatments,irradiating two points(each side of the nasal ala),for 2 min each point at 2 h after exercise.Samples were collected in the beginning of training(week 0),as well as in the end of 2nd,4th and 6th week.Blood cell classification,as well as the content of serum immunoglobulin(IgG and IgM),complement(C3 and C4),cytokine(TNF-α and IL-10),and CRP were measured.Tissue morphology of thymus,bone marrow,and spleen were observed.Results:1)Six-week incremental load training could induce a dynamic and systematic negative change of immune function in rats,involving the number of circulating immune cells,the content of immunoglobulin and complement,the balance of pro-inflammatory and anti-inflammatory cytokine,the proliferation function of T lymphocyte of spleen,and the histological morphology of central and peripheral immune organs.2)Low power laser significantly improved EIS,with the following changes:up-regulated lymphocyte percent,IL-10 content,and the ratio of IL-10/TNF-a;reduced circulating WBC,NE cells and CRP;enhanced proliferation function of T lymphocyte;and improved structure of bone marrow and thymus.3)High power laser up-regulated proliferation function of T lymphocyte,and down-regulated circulating WBC and NE cells.However,CRP and T
