摘 要:目的探讨黄芪甲苷对妊娠期高血压(HDCP)模型大鼠胎盘组织中缺氧诱导因子-1α(HIF-1α)、可溶性血管内皮生长因子受体-1(sFLT-1)和胎盘生长因子(PLGF)表达水平的影响。方法将30只SD孕鼠随机分为对照组、模型组和黄芪组各10只。于妊娠第12天,模型组和黄芪组灌胃亚硝基左旋精氨酸甲酯10 mg/kg,连续7 d,建立大鼠HDCP模型,对照组灌胃等量生理盐水。于妊娠第16天,黄芪组灌胃20 mg/kg黄芪甲苷溶液,模型组和对照组分别灌胃等量的生理盐水,1次/d,连续灌胃6 d。于妊娠第12天、16天、17天及21天测尾动脉血压和24 h尿蛋白含量;处死后,采用ELISA法测定血清中白介素(IL)-6、肿瘤坏死因子(TNF-α)、超氧化物歧化酶(SOD)和丙二醛(MDA)水平,采用实时荧光定量PCR(RT-PCR)和Western Blot胎盘组织中HIF-1α、sFLT-1和PLGF的表达。结果与对照组相比,模型组和黄芪组尾动脉血压、24 h尿蛋白含量、血清IL-6和TNF-α、HIF-1α和sFlt-1蛋白和mRNA表达水平均显著升高(P<0.05),模型组PLGF的蛋白和mRNA表达水平显著下降(P<0.05),黄芪组PLGF的蛋白和mRNA表达水平显著升高(P<0.05);与模型组相比,黄芪组尾动脉血压、24 h尿蛋白含量、血清IL-6和TNF-α、HIF-1α和sFlt-1蛋白和mRNA表达水平均显著下降(P<0.05),PLGF的蛋白和mRNA表达水平显著升高(P<0.05)。结论黄芪甲苷可以使HDCP大鼠的HIF-1α和sFlt-1的表达下调,PLGF的表达上调,对HDCP具有一定的治疗作用。Objective To explore the effects of astragaloside on expression levels of hypoxia-inducible factor-1α(HIF-1α),soluble vascular endothelial growth factor receptor-1(sFLT-1)and placental growth factor(PLGF)in placental tissues of model rats with hypertensive disorder complicating pregnancy(HDCP).Methods 30 SD pregnant rats were randomly divided into control group,model group and Huangqi group,with 10 rats in each group.On the 12th day of pregnancy,Model group and Huangqi group were intragastrically administered with 10 mg/kg of nitroso-L-arginine methyl ester for 7 d,and the rat HDCP model was established.The control group was given the same amount of normal saline.On the 16th day of pregnancy,Huangqi group was intragastrically administered with 20 mg/kg of astragaloside solution,and model group and control group were intragastrically administered with the same amount of normal saline once daily for 6 d.The tail artery blood pressure and 24 h urine protein content were measured on the 12th d,16th d,17th d and 21st d of pregnancy.After sacrifice,the levels of serum interleukin(IL)-6,tumor necrosis factor(TNF-α),superoxide dismutase(SOD)and malondialdehyde(MDA)were determined by ELISA.The real-time fluorescent quantitative PCR(RT-PCR)and Western Blot were used to detect the expression levels of HIF-1α,sFlt-1 and PLGF in placental tissues.Results Compared with control group,the tail artery blood pressure,24 h urine protein content,levels of serum IL-6 and TNF-α,and protein and mRNA expression levels of HIF-1αand sFlt-1 in model group and Huangqi group were significantly increased(P<0.05),and the protein and mRNA expression levels of PLGF in model group were significantly decreased(P<0.05),and the protein and mRNA expression levels of PLGF in Huangqi group were significantly increased(P<0.05).Compared with model group,the tail artery blood pressure,24 h urine protein content,serum IL-6 and TNF-α,and protein and mRNA expression levels of HIF-1αand sFlt-1 were significantly decreased in Huangqi group(P<0.05),and
