丹参提取物及益心舒片中丹酚酸B在大鼠体内的药物代谢动力学研究

摘  要:目的建立反相高效液相色谱法(reversed phase high performance liquid chromatography,RP-HPLC)测定大鼠血浆中丹酚酸B浓度的方法,研究丹参提取物及益心舒片中丹酚酸B在大鼠体内药动学特征.方法大鼠分别灌胃给予丹参提取物、益心舒片,眼眶后静脉丛取血,离心得血浆,加甲醇沉淀蛋白处理后测定丹酚酸B的血药浓度,DAS 2.0药动学软件处理数据.结果丹酚酸B血浆浓度在0.253~10.120μg/ml之间,线性关系良好,检测限为0.063μg/ml,高、中、低浓度样品回收率分别为96.74%、97.51%、98.08%.大鼠灌胃丹参提取物和益心舒片后的药-时曲线均符合二房室模型.灌胃益心舒片后,与灌胃丹参提取物相比,益心舒片中测定的丹酚酸B的Cmax和AUC0→∞结果优于丹参提取物.结论本研究建立的RP-HPLC测定法专属、准确、灵敏,适用于丹参提取物及益心舒片中丹酚酸B的药动学研究.且研究显示,益心舒片中的其他药材配伍可以促进丹酚酸B的吸收,提高丹酚酸B的生物利用度.Objective The objective of this study was to establish a reversed phase high performance liquid chromatography(RP-HPLC)method for the determination of salvianolic acid B in rat plasma,and to study the pharmacokinetic characteristics of salvianolic acid B in Salvia miltiorrhiza extract and Yixinshu tablets in rats.Methods Rats were given Salvia miltiorrhiza extract and Yixinshu tablet by gavage,blood was collected from posterior orbital venous plexus,plasma was centrifuged,salvianolic acid B concentration was determined after treatment with methanol precipitation protein,and data were processed by DAS 2.0 pharmacokinetic software.Results A good linear relationship of salvianolic acid B was obtained between 0.253 and 10.120μg/ml,the limit of determination was 0.063μg/ml.The recovery rates of high,medium and low concentrations were 96.74%,97.51%and 98.08%,respectively.The drug-time curves of rats after oral administration of Salvia miltiorrhiza extract and Yixinshu tablets were consistent with the two-compartment model.After the administration of Yixinshu tablets,the Cmax and AUC0→∞results of salvianolic acid B measured in Yixinshu tablets were better than those of Salvia miltiorrhiza extract.Conclusions The RP-HPLC method is specific,accurate and sensitive.It is suitable for the pharmacokinetic study of salvianolic acid B in Danshen extract and Yixinshu tablets.And studies have shown that the compatibility of other herbs in Yixinshu tablets can promote the absorption of salvianolic acid B and improve the bioavailability of salvianolic acid B.
