摘　　要：目的探讨山楂消脂胶囊对饮食诱导的肥胖小鼠脂代谢紊乱的调节作用及其分子机理。方法高脂饮食建立肥胖小鼠模型,分别以辛伐他丁、山楂消脂胶囊高、中、低剂量进行灌胃治疗30 d,以正常小鼠灌胃同等剂量的生理盐水作为空白对照组。处死后测定各组小鼠的脂肪系数和血清中游离脂肪酸的水平;取肝组织检测LPL、FAS的活性;检测脂肪酸β氧化速率和肝组织脂肪酸β氧化限速酶肉毒碱棕榈酰基转移酶-1(carnitine palmitoyltransferase-1,CPT-1)的表达变化情况;用荧光定量PCR的方法检测肝脏组织中PPARα和PPARγ的表达情况。结果与模型组相比,山楂消脂胶囊各剂量组中脂肪系数和血清中游离脂肪酸的水平均显著降低,FAS和LPL活性、脂肪酸β氧化速率显著升高,CPT-1、PPARα和PPARγ的表达水平均显著上升。结论山楂消脂胶囊能有效改善因高脂饮食引起的肥胖和脂代谢紊乱情况。OBJECTIVE To investigate the regulatory effect and mechanism of Shanzhaxiaozhi capsule on lipid metabolism disorder in obese mice induced by high-fat diet. METHODS Obese mice model was established by high-fat diet feeding,then simvastatin or high,medium,low dose Shanzhaxiaozhi capsule was given by intragastric administration. All mice were sacrificed after 30 days,and serum,liver,visceral adipose were collected,fat index and free fatty acid level were measured respectively. The activity of LPL and FAS,β-oxidation rate and the expression of CPT-1 were also examined. In addition,the expression of PPARα and PPARγ in visceral adipose were determined by real-time PCR. RESULTS Compared with the obese group,fat index and free fatty acid level were decreased,the activity of LPL and FAS were increased,β-oxidation rate and the expression of CPT-1,PPARα and PPARγ were also increased significantly. CONCLUSION Shanzhaxiaozhi capsule can effectively improve obesity and lipid metabolism disorder caused by high-fat diet.
