摘  要:采用马铃薯块茎切片漂浮在药液表面、滤纸吸附药液、海绵吸附药液和保湿培养直接涂药4种方法进行了致病疫霉对甲霜灵抗性测定的初步探索。结果显示,块茎切片不能漂浮在药液表面,滤纸吸附法和海绵吸附法均极易引起切片腐烂,只有块茎切片保湿培养直接涂药法能清晰地区分对照以及不同浓度甲霜灵的处理。切片表面菌丝生长覆盖面积及切片变色面积在对照和不同浓度甲霜灵处理之间有显著差异,能用于评价致病疫霉抗药性的强弱,并明确了该方法中甲霜灵的适宜浓度在5~80μg/mL之间。在此基础上,提出了该方法检测致病疫霉抗药性强弱的评判标准。In order to monitor metalaxyl resistance of Phytophthora infestans more timely and accurately, the experiment was conducted to test the possibility of potato tuber slices for detection. After comparing the tuber slices floatage, filter paper adsorption, sponge adsorption and direct daubing methods, we found that tuber slices could not float on metalaxyl solution surface, and both filter paper adsorption and sponge adsorption methods including control as sterile distilled water resulted in tuber slices rot easily. Only direct daubing metalaxyl solution on tuber slice surface under moisture culture could differentiate control and different treatments. The tuber-slice method could evaluate the resistance degree of P. infestans to metalaxyl, since there were significant differences in mycelia growth and tuber slice discoloration between control and different treatments at different metalaxyl concentrations. In addition, the appropriate concentrations of metalaxyl solution were optimized as 5-80 μg/mL. To utilize this method, we also proposed the criteria for resistance evaluation. The establishment of this method can provide a new alternative for timely and accurately detecting metalaxyl resistance of P.infestans in different potato production areas in future.
