摘 要:目的:了解免疫散射比浊法与免疫荧光定量法对于检测尿微量白蛋白的精密度,并以免疫散射比浊法为参考方法判定免疫荧光定量法的系统误差可否接受。方法:采用重复性试验测量精密度:重复测定低、中、高3个浓度水平的样本各20次,共测5d,每天4次;采用方法对比试验测量系统误差:取40个样本(30个0~20mg/L,10个20~200mg/L),正序、倒序各测一次,绘制散点图,用直线回归分析法计算系统误差。结果:2种方法检测结果的变异系数都<7.5%;以散射比浊法为参考方法,荧光定量法检测的系统误差<1/2TEa,且R2≥0.95。结论:免疫散射比浊法和免疫荧光定量法检测尿微量白蛋白的精密度良好,均可运用于临床。Objective To detect the precision of scattering immunoturbidimetry and immunofluorescence quantitative method in the determination of urinary microalbumin, and to examine the system error of the immunofluorescence quantitative method with scattering immunoturbidimetry used as the referential method. Methods The repeatability test was used to measure the precision: 20 repetitions of the samples in the lower, middle and high levels were measured 4 times a day within 5 days. The system error was detected by methods comparison test: 40 samples (30 in 0-20 mg/L, 10 in 20-200 mg/L) were measured in direct sequence and reverse sequence separately, and scatter diagram and linear regression analysis were used to calculate the system error. Results The coefficients of variation of the two methods were less than 7.5%. The systematic error of fluorescence quantitative method was less than 1/2TEa and R2 was not lower than 0.95. Conclusion Scattering immunotu-rbidimetry and immunofluorescence quantitative method gain high precision when used to detect urinary microalbumin, and thus are worthy promoting clinically.
