狼毒大戟提取物及其单体化合物对人乳腺癌细胞株MDA-MB-231增殖和周期影响

摘　　要：目的:观察狼毒大戟提取物及其单体化合物对人乳腺癌细胞株MDA-MB-231增殖及周期影响。方法:将狼毒大戟总提取物、狼毒素、狼毒素A、狼毒素C分别作用于MDA-MB-231细胞株,用CCK-8法检测狼毒大戟总提取物对MDA-MB-231增殖影响,采用药物浓度为30mg/mL、50mg/mL、100mg/mL、300mg/mL、500mg/mL、1000mg/mL。用CCK-8法检测狼毒素、狼毒素A、狼毒素C对MDA-MB-231增殖影响,药物浓度均采用25mg/mL、50mg/mL、75mg/mL、100mg/mL、120mg/mL;采用流式检测法,检测狼毒大戟总提取物对MDA-MB-231周期影响。结果:狼毒大戟提取物、狼毒素、狼毒素A、狼毒素C对MDA-MBA-231细胞株增殖影响IC50分别为486.69mg/mL、80.42mg/mL、62.49mg/mL、98.64mg/mL。狼毒大戟提取物可使MDA-MB-231细胞株的细胞周期S期上升、G2/M、G0/G1期均下降。结论:狼毒大戟总提取物及其单体化合物狼毒素A、狼毒素、狼毒素C对MDA-MB-231细胞株增殖均有抑制作用,其中狼毒素A对细胞增殖抑制率最高。狼毒大戟提取物可使三种细胞株的细胞周期均阻滞于S期。Objective:To study the effects of extracts from Euphorbia fischeriana Steud.and its monomeric compounds on proliferation and cell cycle of human breast cancer cell line MDA-MB-231.Methods:Total extracts of Euphorbia fischeriana Steud,Chamaejasmenin,Chamaejasmenin A and Chamaejasmenin C were applied to MDAMB-231 cells respectively,and the total extract of Stellera chamaejasme L.was detected by CCK-8 method proliferation effects,using drug concentrations of 10 mg/mL,50 mg/mL,100 mg/mL,300 mg/mL,500 mg/mL,1000 mg/mL.The proliferation of MDA-MB-231 was detected by CCK-8 method,and the drug concentration was 25 mg/mL,50 mg/mL,75 mg/mL,100 mg/mL,120 mg/mL;Flow cytometry was used to detect the effect of total extract of Stellera chamaejasme on MDA-MB-231 cycle.Results:he IC50 of the effects of extracts of Stellera chamaejasme,C.hamaejasmenin,Chamaejasmenin A and Chamaejasmenin C on MDA-MBA-231 cell line were 486.69 mg/ml,IC50 80.42 mg/mL,62.49 mg/mL,98.64 mg/mL,respectively./ml.The extract of Stellera chamaejasme L.increased the cell cycle S phase of MDA-MB-231 cell line,and decreased G2/M and G0/G1 phase(P<0.01,statistically significant).Conclusion:The total extract of Stellera chamaejasme L.and its monomeric compounds,Chamaejasmenin A,Chamaejasmenin and Chamaejasmenin C,inhibited the proliferation of MDA-MB-231 cells.Chamaejasmenin A had the highest inhibition rate on cell proliferation.The extract of Stellera chamaejasme can arrest the cell cycle of the three cell lines in the S phase.
