摘  要:目的探讨白细胞介素-6(IL-6)对胰腺癌BxPC-3细胞增殖的影响及其调控Toll受体4(TLR4)/核转录因子kappaB(NF-κB)信号通路作用机制。方法 MTT法检测IL-6对BxPC-3细胞增殖的影响并计算增殖率;台盼蓝拒染法检测IL-6对BxPC-3细胞生长的影响并绘制生长曲线;克隆形成实验观察IL-6对BxPC-3细胞克隆形成的影响并计算克隆形成率;逆转录聚合酶链反应(RT-PCR)法检测IL-6对BxPC-3细胞TLR4、髓样化因子(MyD88)、NF-κB、白细胞介素-1β(IL-1β)及肿瘤坏死因子-α(TNF-α)mRNA表达水平的影响;Westen blot法检测IL-6对BxPC-3细胞TLR4、MyD88、NF-κB、IL-1β及TNF-α蛋白表达水平的影响。结果与阴性对照组比较,IL-6可促进BxPC-3胰腺癌细胞生长,升高细胞增值率和克隆形成率;IL-6干预后,BxPC-3胰腺癌细胞TLR4、MyD88、NF-κB、IL-1β和TNF-α等因子的mRNA和蛋白表达水平均明显上调。结论 IL-6可促进BxPC-3胰腺癌细胞生长增殖,其机制可能与上调TLR4/NF-κB信号通路有关。Objective To explore the effects of IL-6 on the proliferation of human pancreatic cancer BxPC-3 cells and the TLR4/NF-κB signaling pathway-related mechanism.Methods MTT assay was used to detect the effects of IL-6 on the proliferation of BxPC-3 cells and the cell proliferation rate was calculated.Trypan blue staining assay was used to count the alive cells and draw the cell growth curve.Colony formation assay was used to observe the effects of IL-6 on the BxPC-3 cell clone formation and the colony formation rate was calculated.The expression levels of TLR4,MyD88,NF-κB,IL-1β and TNF-α mRNA and protein in BxPC-3 cells were detected by reverse transcription-polymerase chain reaction(RT-PCR)and Western blot analysis.Results Compared with the negative control group,IL-6 could promote the proliferation of the human pancreatic cancer BxPC-3 cell,and increase its proliferation rate and clone formation rate.After the treatment with IL-6,the mRNA and protein expression levels of TLR4,MyD88,NF-kappa B,IL-1 beta and TNF-alpha in BxPC-3 pancreatic cancer cells were significantly up-regulated.Conclusion IL-6 can promote the human pancreatic cancer BxPC-3 cells proliferation,and its mechanism may be related to up-regulation of TLR4/NF-κB signaling pathway.
