全蝎白术白头翁混合发酵物超声水提工艺的优化及体外抗肿瘤活性研究

摘　　要：目的优选全蝎白术白头翁混合发酵物(MFSAP)的超声水提工艺并进行体外抗肿瘤活性的初步研究。方法通过球孢白僵菌双向固体发酵技术制备MFSAP,以水溶性总蛋白及总多糖的含量为指标,在单因素试验的基础上,采用L9(3^4)正交表考察溶剂用量、超声时间、pH值对超声水提工艺的影响,并研究最佳水提工艺制备的MFSAP对乳腺癌细胞株MCF-7的抑制作用。结果MFSAP的最佳水提工艺为:溶剂用量40倍,超声时间60min,pH11。在此条件下,水提液中水溶性总蛋白的含量为226.78mg·g^-1(RSD=0.42%),水溶性总多糖的含量为222.99mg·g^-1(RSD=0.35%),综合评分为2.30(RSD=3.40%);MFSAP具有较好的体外抗肿瘤效果。结论所用工艺优化方法合理可行,所得结果稳定可靠;球孢白僵菌双向固体发酵技术可提高此中药复方的抗肿瘤活性。OBJECTIVE To optimize the ultrasonic water extraction of mixed fermentation of Scorpio, Atractylodes and Pulsatilla( MFSAP),and to study its antitumor activity in vitro. METHODS MFSAP was prepared by the bidirectional solid fermentation technology of Beauveria bassiana. With the content of total water-soluble protein and polysaccharide as indexes, the amount of solvents, ultrasonic time, and pH were investigated by L9(3^4 ) orthogonal table on the basis of Single factor experiment. The inhibitory effect of the best water extract using the extraction technique on mammary cancer cells strain MCF - 7 was studied. RESULTS The optimum conditions for water extraction of MFSAP were as follows: 40-folds of water amount, ultrasonic time of 60 min, and pH at 11. Under these conditions, the content of total water - soluble protein was 226. 78 mg·g^-1 (RSD =0.42%),total water-soluble polysaccharide was 222. 99 mg·g^-1 ( RSD =0. 35%),the comprehensive score was 2. 30 (RSD =3. 40%). And MFSAP had preferable antitumor effect in vitro. CONCLUSION The process optimization method is reasonable and feasible, and the results are stable and reliable. The bidirectional solid fermentation technology of Beauveria bassiana could improve the antitumor activity of the compound Chinese medicine.
