摘 要:目的优化灵芝子实体中灵芝酸类成分的亚临界水提取工艺。方法采用单因素实验设计,HPLC法测定以灵芝酸A和灵芝酸C2为代表的灵芝酸的提取量,考察提取温度、时间、液料比、提取次数对灵芝酸类成分提取量的影响。结果优化的灵芝酸亚临界水提取工艺为:液料比为11,于140℃、5MPa条件下提取5min。结论亚临界水提取法与索氏提取法的提取量相当,较超声提取法高,但耗时更短,且不需要有机溶剂,有利于保护环境。OBJECTIVE To optimize the subcritical water extraction ( SWE) technology of ganoderic acids from the fruiting body of Ganoderma lucidum. METHODS The effect of extraction parameters including temperature, extraction time, the solvent/sample ratio and extraction frequency on the yields were investigated by Single - factor experiment design. The contents of ganoderic acid A and ganoderic acid C2 as the index were analyzed by HPLC. RESULTS The optimum SWE extraction conditions for ganoderic acids were as follows: G. lucidum sample was extracted once at the solvent/sample ratio of 11 under 5 MPa and 140℃ for 5 minutes. CONCLUSION The extraction rate of SWE is equivalent to Soxhlet extraction and better than that of Ultrasonic extraction, but SWE requires less handling time and no organic solvent consumption, which was beneficial to environmental protection.
