泽兰水提物调节PKC同工酶治疗早期糖尿病肾病的研究

摘　　要：目的:探讨泽兰水提物对早期糖尿病肾病(DKD)的作用机制。方法:随机将SD大鼠分成正常组、模型组、泽兰水提物低、高剂量组和氯沙坦组,予高脂饲料喂养、链脲佐菌素腹腔注射建立早期糖尿病肾病模型,分别予泽兰水提物、氯沙坦灌胃,检测血、尿生化指标,QPCR及westernblot法分别检测PKC同工酶mRNA、蛋白表达水平。结果:泽兰水提物可以减少尿微量白蛋白/尿肌酐比值(UACR),效果与氯沙坦相当。相较于氯沙坦,泽兰水提物无增加肾损伤的风险。泽兰水提物对PKCα、PKCβⅠ的抑制作用类似于氯沙坦。模型组大鼠肾组织的PKCε较正常组显著降低。结论:泽兰水提物可以抑制PKCα、PKCβⅠ,减轻早期DKD蛋白尿,泽兰水提物不影响早期DKD肾功能。不同于PKCα、PKCβⅠ,PKCε在DKD中可能起到保护作用。Objective: To examine preliminarily the effect and mechanism of the aqueous extract of lycopus on early stage of diabetic kidney disease(DKD). Methods: SD rats were randomly divided into normal group,model group, low-dose and high-dose groups of lycopus water extract and losartan group. Early diabetic nephropathy model was established by feeding high-fat diet and intraperitoneal injection of streptozotocin. The low-dose and high-dose groups of lycopus water extract and losartan groups were given lycopus water extract and losartan respectively by gavage. The biochemical indexes of blood and urine were detected, and the levels of PKC isozyme mRNA and protein expression were detected by QPCR and Western-blot respectively. Results: The aqueous extract of lycopus could reduce the urinary microalbumin/creatinine ratio(UACR), and the effect was similar to that of losartan. Compared with losartan, aqueous extract of lycopus did not increase the risk of kidney injury. The inhibitory effect of aqueous extract of lycopus on PKC α and PKC βⅠ was similar to that of losartan. The PKC epsilon in kidney tissue of model group rats was significantly lower than that of normal rats.Conclusions: The aqueous extract of lycopus can inhibit PKC α and PKC βⅠ, and alleviate early DKD proteinuria. The aqueous extract of lycopus does not affect early DKD renal function. Unlike PKC α and PKC β Ⅰ,PKC ε may play a protective role in DKD.
