摘 要:宫颈癌的发生发展是一个多基因调控的复杂过程,微小RNA(microRNA,miRNA/miR)-504和6-磷酸葡萄糖脱氢酶(Glucose-6-phosphate dehydrogenase,G6PD)在宫颈癌中的表达及其与高危型人乳头瘤病毒(Human papilloma virus,HPV)感染的关系尚未见报道。本研究通过检测miR-504、G6PD在宫颈癌中的表达水平,研究其表达水平与高危型HPV感染的关系。收集2016年5月至2017年8月在石家庄市第四医院行宫颈癌手术切除患者46例为研究对象,取宫颈癌组织及癌旁组织,荧光定量PCR检测宫颈组织中miR-504和G6PD的mRNA表达水平,免疫组化检测G6PD在宫颈组织中的蛋白表达水平,采用HC2技术检测宫颈组织中HPV感染情况。结果显示宫颈癌组织中miR-504表达水平显著低于癌旁组织(P <0.05);宫颈癌组G6PD mRNA表达水平显著高于癌旁组织(P<0.05);免疫组化结果显示宫颈癌组G6PD阳性表达率显著高于癌旁组织(P<0.05);miR-504表达水平与HPV感染呈负相关(r=-0.773,P<0.05),G6PD表达强度与HPV感染呈正相关(r=0.669,P<0.05);miR-504表达水平与G6PD mRNA表达水平呈负相关(r=-0.740,P<0.05)。本研究提示miR-504在宫颈癌组织中表达下调,与HPV感染呈负相关;G6PD在宫颈癌组织中表达上调,与HPV感染呈正相关;miR-504和G6PD mRNA表达水平呈负相关,推测miR-504可能负调控G6PD参与HPV感染患者宫颈病变发展过程。The occurrence and development of cervical cancer is a complex process of multi-gene regulation.The expression of microRNA(miR)-504 and glucose 6-phosphate dehydrogenase(G6 PD)in cervical cancer and their relationship with high-risk human papillomavirus(HPV)infection have not been reported. To detect the expression levels of miR-504 and G6 PD in cervical cancer,and explore their relationships with high-risk HPV infection. Fourty-six patients with cervical cancer who underwent surgical resection in the Fourth Hospital of Shijiazhuang City,China from May 2016 to August 2017 were selected as subjects.Cervical cancer tissues and adjacent tissues were taken. The expression levels of miR-504 and G6 PD in cervical tissues were detected by fluorescence quantitative PCR. The expression of G6 PD in cervical tissues was detected by immunohistochemistry. HPV infection in cervical tissues was detected by HC2. The results showed that the expression of microRNA-504 in cervical cancer tissues was significantly lower than that in adjacent tissues(P<0.05). The expression of G6 PD in cervical cancer tissues was significantly higher than that in adjacent tissues(P<0.05). Immunohistochemical results showed that the expression rate of G6 PD in cervical cancer group was significantly higher than that in adjacent tissues(P<0.05).The expression level of miR-504 was negatively correlated with HPV infection(r=-0.773,P<0.05),and the expression intensity of G6 PD was positively correlated with HPV infection(r= 0.669,P<0.05);the expression level of microRNA-504 was negatively correlated with that of G6 PD(r=-0.740,P<0.05). The present study suggest that the expression of miR-504 is down-regulated in cervical lesions,it is negatively correlated with HPV infection.The expression of G6 PD is up-regulated in cervical lesions,it is positively correlated with HPV infection.There is a negative correlation between the expressions of miR-504 with G6 PD mRNA.It is speculated that miR-504 may be involved in the development of cervical lesions in HPV inf
