摘  要:建立了一种基于鲁米诺(luminol)化学发光的流动注射-化学发光系统,考察了蠕动泵流速、luminol浓度等对系统发光强度的影响,确定了最佳实验参数,并采用该系统对BSA的构象变化活性进行研究。结果表明,BSA在碱性环境中可显著增强luminol的发光强度,华法林对于BSA/luminol体系发光强度的抑制作用在一定浓度范围内(0.1~1600μmol·L^-1)呈现良好的线性关系。该方法稳定性高,结果可靠且易于操作,可为其他功能蛋白质构象变化活性表征提供方法学的借鉴。In the current work, the flow injection-chemiluminescence (FL-CL) system was successfully established. To obtain the optimal experimental parameters, the flow rate, the length of mixing tube and the concentration of luminol and BSA were tested. Taking bovine serum albumin (BSA) as an example, the conformational change was investigated by the proposed FL-CL method. The result showed that BSA could enhance the intensity of the FI-CL system under alkaline condition. Meanwhile, a good linear relationship was found between the loss of CL intensity and the concentration of wafarin, and the concentration range of wafarin was 0.1~1600 μmol/L. The proposed FI-CL method was believed to preserve the advantages of high stability and reliable results. It would be an alternative in characterizing the conformational change of other functional proteins.
