摘 要:生长素输出载体蛋白PIN-formed(PIN)是一类调控植物生长素极性运输的重要载体元件,与植物生长发育密切相关。利用生物信息学方法对百脉根(Lotuscorniculatus)PIN基因家族成员(LjPIN)进行筛选、鉴定,并对其结构特征、系统进化、编码蛋白质性质、顺式作用元件组成和表达特性等进行研究,为其功能鉴定及利用奠定基础。结果表明,百脉根基因组中含有27个LjPIN成员,主要分布在1-5号染色体上;27个LjPIN基因含8个可变剪切位点,编码35个蛋白质;除了LjPIN4、LjPIN13、LjPIN23基因外,其他LjPIN基因均含有内含子,内含子数和外显子数均为1~10个;LjPIN与拟南芥AtPIN、大豆GmPIN在进化关系上较接近,与水稻OsPIN较远;LjPIN蛋白大多为碱性的两性稳定蛋白质,一般含有5种保守基序,基序数量2~9个;LjPIN基因除了含有顺式作用基本元件CAAT-box和TATA-box外,还含有光响应、激素响应及生长发育相关的调控元件;11个LjPIN基因在根、茎、叶、花和不同形成时期种子中呈时空差异表达。PIN-formed protein (PIN) is an important carrier regulating the auxin's polar transport,and is closely related to the plant growth and development.In this study,the PIN family genes ( LjPIN ) were identified from the genetics databases of Lotus corniculatus.These genes were further analyzed for the gene construction,evolution relationship,protein features,cis-acting element site and tissue expression characteristics,etc..Results showed that 27 LjPIN gene members encoding 35 proteins by eight alternative splicing sites were found mainly in chromosomes 1-5.Most genes were made up of 1-10 introns and exons except for the LjPIN4,LjPIN13 and LjPIN23.Evolutionary relationship of LjPIN with Arabidopsis AtPIN and soybean GmPIN was closer than with rice OsPIN.Most LjPIN proteins with 5 kinds of conserved motifs (its number was 2-9) were alkaline and stable.The genes not only included cis-acting elements of CAAT-box and TATA-box,but also contained light-responsive,hormone-responsive as well as growth-related regulatory elements.11 differential expression genes were discovered in different developmental stages and various tissues,such as root,stem,leaf,flower and seed.
