摘 要:目的研究奶蓟草和疏叶骆驼刺对小鼠酒精性肝损伤的保护机制。方法将90只小鼠随机分为9组,即:正常组,模型组,联苯双酯组,奶蓟草高、中、低剂量组,疏叶骆驼刺高、中、低剂量组。除正常组外,其他各组采用酒精度为55度的白酒连续灌胃4周,之后,联苯双酯组、奶蓟草组和疏叶骆驼刺组分别给与对应药物治疗4周,然后测定各组小鼠的相关生化指标,如ALT、AST、TC、TG、TNF-α、NF-κB和TNF-αmRNA的表达的含量。结果模型组小鼠血清中的ALT和AST含量明显高于正常组;与模型组相比,奶蓟草和疏叶骆驼刺高剂量组均可明显降低血清中ALT、AST、TC、TG的含量,并且明显增加肝匀浆中SOD和GSH的含量而明显降低MDA含量;奶蓟草和疏叶骆驼刺高剂量组可明显降低TNF-α和NF-κB的含量;奶蓟草和疏叶骆驼刺高、中剂量组可显著降低小鼠肝组织中TNF-αmRNA的表达的含量。结论奶蓟草和疏叶骆驼刺对酒精性肝损伤小鼠具有较好的保护作用。This paper aims to evaluate the protective effect of Silybum marianum and Alhagi sparsifolia on alcohol-induced acute liver injury in mice by observing serum biochemical indexes, inflammatory cytokine release,liver antioxidant capacity, and changes of histopathological damage in mice with alcoholic liver injury. Total of 90 ICR mice were recruited and randomly divided into nine groups: control group, model group, biphenyl dimethylester group, Silybum marianum low, moderate and high dose groups, Alhagi sparsifolia low, moderate and high dose groups. The levels of AST, ALT, TC and TG in serum were measured, the levels of TNF-α, MDA, SOD and GSH in liver were detected, and the relative expression of TNF-α mRNA in liver tissue was measured by the RT-PCT.Furthermore, the histopathological change of liver was observed. Data showed that the levels of ALT and AST in serum of model group were significantly higher than those of normal group. Compared with the model group,the high dose groups of Silybum marianum or Alhagi sparsifolia demonstrated significant lower levels of ALT, AST, TC and TG in serum, but significant higher levels of SOD and GSH in liver homogenate, resulting in decrease of MDA level. High dose of Silybum marianum or Alhagi sparsifolia significantly reduced the levels of TNF-α and NF-κB. Furthermore, the relative expression of TNF-α mRNA in liver tissue of mice was significantly decreased in high and medium dose groups of Silybum marianum or Alhagi sparsifolia. Therefore, the Silybum marianum and Alhagi sparsifolia has certain protective effect on acute alcohol-induced liver injury.
