摘  要:目的通过建立异基因骨髓移植(allogeneic bone marrow transplantation,allo-BMT)小鼠模型联合腹腔给药,探讨金黄色葡萄球菌肠毒素C(staphylococcal enterotoxin C,SEC)对allo-BMT小鼠的T细胞早期重建的影响。方法以BALB/C雄性小鼠为受鼠,C57BL/6雄性小鼠为供鼠,在移植前12 h对受鼠进行全身性照射的预处理,进行异基因骨髓内骨髓移植(intra-bone marrow bone marrow transplantation,IBM-BMT)后将其随机分为两组腹腔给药。实验组给予SEC 100μL/(g·d),对照组给予同剂量的生理盐水,每组10只。每天检测受鼠的体质量变化情况,并于移植2周后应用流式细胞仪检测两组外周血及脾脏T淋巴细胞重建情况,采用ELISA检测血浆IFN-γ、IL-2、TNF-α的分泌水平。结果实验组、对照组外周血CD8^+T细胞比例(12.63±1.33)%、(6.67±0.53)%,组间差异有统计学意义(P <0.01)。实验组脾脏和外周血的CD4^+T细胞比例分别为(1.798±0.110)%、(1.470±0.075)%,高于对照组(1.418±0.069)%、(1.112±0.064)%,差异有统计学意义(P <0.05)。实验组血浆中分泌的细胞因子IFN-γ、IL-2和TNF-α水平分别为(383.7±49.22)pg/mL、(17.49±1.434)pg/mL和(144.9±14.38)pg/mL,高于对照组(230.0±13.17)pg/mL、(12.38±0.759)pg/mL和(102.0±7.58)pg/mL,组间差异有统计学意义(P <0.05)。结论腹腔注射SEC能有效促进移植后T细胞早期重建。Objective To explore the effects of intraperitoneal administration of staphylococcal enterotoxin C on T cells reconstitution after allogeneic bone marrow transplantation(allo-BMT). Methods One day before transplantation the recipient BALB/C mice were given total-body irradiation,then transplanted with bone marrow cells from donor C57BL/6 mice and randomly divided into two groups. The recipient mice transplanted with BMT plus SEC 100 μL/(g·d)were served as experimental group,and those mice transplanted with BMT plus intraperitoneal injection of the same amount of normal saline were served as control group(n = 10 each).The changes of weight about two groups were measured every day after the intraperitoneal injection of SEC. Flow cytometry was used to examine the T cells subsets in peripheral blood and spleen of recipient mice 2 weeks after bone marrow transplantation. Meanwhile,the serums were collected at the same time to detect the levels of IFN-γ,IL-2 and TNF-α by ELISA. Results Percentage of CD8^+T cells in experimental group and control group was(12.63±1.33)% and(6.67 ± 0.53)% respectively,and there was significant difference between pairwise comparisons(P < 0.05).Percentage of CD4^+T cells of the spleen and the peripheral blood in experiment group(1.798 ± 0.110)% and(1.470 ± 0.075)% was significantly higher than control group(1.418 ± 0.069)% and(1.112 ± 0.064)%(P < 0.05).The levels of IFN-γ,IL-2 and TNF-α in experiment group(383.7 ± 49.22)pg/mL,(17.49 ± 1.434)pg/mL and(144.9 ± 14.38)pg/mL was significantly higher than that in control group(230.0 ± 13.17)pg/mL,(12.38 ± 0.759)pg/mL and(102.0 ± 7.58)pg/mL(P < 0.05). Conclusion Intraperitoneal administration of staphylococcal enterotoxin C can effectively promote the early reconstitution of T lymphocytes in the recipient mouse after allo-BMT.
