蒙药童格勒格-1对急性辐射损伤小鼠保护作用的研究

摘  要:目的:观察蒙药童格勒格-1对X射线辐射损伤小鼠的保护作用。方法:将40只小鼠随机分为空白组、辐射组、童格勒格-1组和童格勒格-1辐射组,每组10只。其中空白组和辐射组给予蒸馏水,童格勒格-1组和童格勒格-1辐射组给以童格勒格-1提取物,连续灌胃15d后,对辐射组和童格勒格-1辐射组小鼠进行1.5GyX射线辐射,24h后采集附睾应用伊红染色法检测小鼠精子畸形率;采集骨髓组织,应用吉姆萨染色法检测小鼠骨髓嗜多染红细胞的微核率,应用Annexin-V-FITC/PI染色检测小鼠骨髓幼稚红细胞的凋亡率。结果:辐射组小鼠精子畸形率、嗜多染红细胞微核率、细胞凋亡率显著高于空白组(P<0.05),而童格勒格-1组与空白组差异不显著(P>0.05);与辐射组相比,童格勒格-1辐射组的精子畸形率、嗜多染红细胞微核率、骨髓幼稚红细胞凋亡率则显著降低(P<0.05)。结论:蒙药童格乐格-1对X射线造成的损伤具有保护作用。Objective:To study the protective effect of Mongolian medicine TGLG-1 on X-ray radiation-injured mice.Methods:40 mice were divided into the blank group,the radiation group,the TGLG-1 group and the TGLG-1 radiation group,ten in each group.The blank group and the radiation group were given distilled water,while the TGLG-1 group and the TGLG-1 radiation group were given TGLG-1 for 15 days before 1.5 Gy X-ray irradiation.The sperm deformity was detected by eosin staining;Giemsa's staining was adopted to detect the micronucleus rate of bone marrow polychromatic erythrocyte;the apoptotic rate of myelocyte was detected by Annexin V-FITC/PI staining.Results:The sperm abnormality rate,the micronucleus rate and the apoptotic rate in the radiation group were significantly higher than those in the control group(P<0.05).There was no significant difference between the blank group and the TGLG-1 group.Compared with the radiation group,the sperm abnormality rate,the micronucleus rate of polychromatic erythrocytes and the apoptotic rate of myelocyte in the TGLG-1 radiation group were significantly reduced(P<0.05).Conclusion:Mongolian medicine TGLG-1 has a radiation protective effect on X-ray radiation-injured mice.
