健脾益肠散对LPS诱导树突状细胞成熟标志CD83表达的影响

摘　　要：目的观察健脾益肠散对体外脂多糖(LPS)诱导的树突状细胞(DC)成熟标志CD83表达的影响。方法原代分离、培养C57BL/6小鼠DC细胞,采用流式细胞术对DC细胞的主要标记物CDla进行鉴定,LPS诱导细胞构建DC细胞成熟模型。将培养的DC细胞随机分为空白对照组、模型组、中药含药血清组、空白血清组和IL-10组。干预24 h后,采用Western blotting、RT-PCR技术分别检测各组DC细胞中CD83的蛋白和mRNA表达。结果与空白对照组比较,模型组DC细胞CD83的蛋白和mRNA表达均明显增多(P <0.05或P <0.01);中药含药血清组DC细胞CD83的蛋白和m RNA表达减少,与模型组和空白血清组比较差异均有统计学意义(P<0.05)。结论健脾益肠散对LPS诱导的DC细胞成熟标志CD83的蛋白和mRNA表达具有明显的抑制作用,可能是其发挥免疫耐受作用治疗溃疡性结肠炎(UC)的重要机制。Objective:To investigate the effect of Jianpi Yichang Powder on the expression of mature marker CD83 in DC induced by LPS in vitro. Methods:C57BL/6 mouse DC were primitively isolated and cultured. The main marker CDla in DC was identified by flow cytometry. Establishment of DC maturation model was induced by LPS. The cultured DC were randomly divided into the blank control group,the model group,TCM group,serum group and IL-10 group. After 24 hours of intervention,the protein and mRNA expression of CD83 in DC were detected by Western blotting and RT-PCR. Results:Compared with the blank control group,the protein and mRNA expression of CD83 in DC were significantly increased in the model group(P < 0.05 or P < 0.01). Those in TCM group were significantly decreased;there was significant difference among the model group,serum group and TCM group(P < 0.05). Conclusion:Jianpi Yichang Powder can significantly inhibit the expression of CD83 and mRNA in DC induced by LPS,which may be an important mechanism of its immune tolerance in the treatment of UC.
