摘  要:目的观察不同剂量温阳健脑方对血管性认知障碍(VCI)模型大鼠血清丙二醛(MDA)、超氧化物歧化酶(SOD)和脑组织多巴胺(DA)的影响,探讨其对VCI大鼠脑保护作用的机制。方法将30只SD大鼠按照改良2VO法制备VCI模型,按照随机数字表法分为空白组、模型组、吡拉西坦组及温阳健脑+吡拉西坦组、温阳健脑低剂量组、温阳健脑高剂量组,每组各6只。造模成功后连续喂药14 d,检测血清MDA、SOD和脑组织DA的含量。结果与空白对照组比较,模型组血清MDA较空白对照组显著升高(P <0.05);模型组血清SOD及脑组织DA较空白对照组显著下降(P <0.05或P <0.01)。与模型组比较,各给药组血清MDA下降,其中温阳健脑高剂量组血清下降最为显著(P <0.01);各给药组血清SOD及脑组织DA均上升,其中温阳健脑高剂量组血清SOD上升最显著(P <0.05或P <0.01)。结论温阳健脑方尤其高剂量组对VCI模型大鼠具有脑保护作用,其机制可能与通过降低血清MDA的含量,提高血清SOD和脑组织DA的含量有关。Objective:To investigate the effects of different doses of Wenyang Jiannao Decoction on serum MDA,SOD and brain tissue DA in rats with vascular cognitive impairment(VCI)and its protective effect on the brain. Methods:VCI model of 30 SD rats was prepared with modified 2 VO method,and were randomly divided into the blank group,the model group,piracetam group,Wenyang Jiannao+piracetam group,Wenyang Jiannao low dose group and Wenyang Jiannao high dose group,6 rats in each. Serum MDA,SOD and brain DA levels were measured after 14 days of continuous administration. Results:Compared with the blank control group,serum MDA in the model group was significantly higher(P < 0.05);serum SOD and brain DA were significantly lower(P <0.05 or P < 0.01). Compared with the model group,the serum MDA decreased in each group,especially in high dose group(P < 0.01);the serum SOD and brain DA increased in each group,and the serum SOD increased most significantly in high dose group(P < 0.05 or P < 0.01). Conclusion:Wenyang Jiannao Decoction,especially high dose,has a brain protective effect on VCI model rats. Its mechanism may be related to the decrease of serum MDA content and the increase of serum SOD and brain DA content.
