摘  要:目的:探讨黄连素对痰湿型多囊卵巢综合征(PCOS)大鼠的治疗作用及疗效机制。方法:雌性SD大鼠颈后皮下注射INS+HCG构建痰湿型PCOS大鼠模型。PCOS模型大鼠随机均分为模型组和黄连素组(n=10),另选取10只正常SD大鼠作为空白组,黄连素组予黄连素2次/d灌胃给药,空白组及模型组予等体积生理盐水灌胃,连续30 d。观察动情周期,间期取材称重,检测血清甾体激素、糖脂代谢水平及糖耐量试验,RT-PCR法测定卵巢CYP17和CYP19mRNA表达及子宫Glut4 mRNA表达水平。结果:(1)与空白组比较,模型组大鼠动情周期明显延长,体质量、卵巢、子宫和性腺旁脂肪重量明显增加,血清甾体激素LH、T、E2水平及LH/FSH比值明显升高,脂代谢指标TG、LDL水平明显升高,OGTT结果显示各时段血糖、FINS水平、HOMA-IR指数均显著增高;(2)与模型组比较,黄连素组大鼠动情周期明显缩短,大鼠体质量、卵巢、子宫和性腺旁脂肪重量明显降低,血清中LH、T、E2水平及LH/FSH比值明显降低,血清TG、LDL水平明显降低,OGTT结果显示各时段血糖、FINS水平、HOMA-IR指数均明显降低。(3)与空白组比较,模型组大鼠卵巢中CYP17a1 mRNA表达明显升高,CYP19a1mRNA表达明显降低,子宫中Glut4mRNA表达明显降低;与模型组比较,黄连素组大鼠卵巢中CYP17a1 mRNA表达明显降低,CYP19a1mRNA表达明显升高,子宫中Glut4mRNA表达明显升高。结论:黄连素能够改善痰湿型PCOS模型大鼠糖脂代谢异常,调控血清甾体激素水平,起到治疗PCOS生殖障碍与代系异常的作用;其改善生殖障碍的分子机制为上调卵巢CYP19a1基因及子宫Glut4基因的表达,下调卵巢CYP17a1基因的表达。Objective:To investigate the therapeutic effect and mechanism of berberine on phlegm-damp PCOS rats. Methods: The female SD rats model of phlegm-damp type PCOS were constructed by subcutaneous injection of INS+HCG.PCOS model rats were randomly divided into model group and berberine group(n=10). Ten normal SD rats were selected as blank group. The berberine group administered berberine twice a day for intragastric administration, and the blank group and model group were intragastrically administrated with equal volume of normal saline twice a day for 30 days.To observe the estrous cycle,the samples were collected at intervals,and we recorded the weight.The serum steroid hormone,glucose lipid metabolism level and OGTT were measured. The expressions of CYP17 and CYP19 mRNA in ovary and Glut4 mRNA in uterus were detected by RT-PCR.Results:(1)Compared with the blank group, the estrus cycle of the model group was significantly prolonged, and the body weight, ovary, uterusand paragonnal fat weights were significantly increased. Serum steroid hormones LH, T and E2 levels and LH/FSH ratio increased significantlyand lipid metabolic indicators TG and LDL levels increased significantly. OGTT results showed that blood glucose, FINS levels and HOMA-IR index were significantly higher in various periods.(2)Compared with the model group, the estrus cycle of the berberine group was significantly shortened, and body weight, ovary, uterusand paragonal fat weights were significantly lower in rats. Compared with the blank group, serum TG, LDL, LH, T and E2 levels and LH/FSH ratio were significantly higher in the model group. Compared with the model group, serum TG, LDL, LH, T and E2 levels and LH/FSH ratio were significantly lower in the berberine group. TG and LDL levels significantly decreased. The blood glucose, FINS levels, HOMA-IR indexand serum TG and LDL levels in the OGTT at each time point decreased in the model group.
