摘 要:目的:基于网络药理学方法,对四妙丸治疗类风湿关节炎的作用机制进行探讨和研究。方法:通过中药系统药理学数据库和分析平台以口服生物利用度和类药性为限定条件,对四妙丸所含化学成分进行筛选并预测作用靶点;通过GeneCards数据库检索类风湿关节炎发病相关的靶点,将四妙丸预测靶点与类风湿关节炎相关靶点导入VENNY 2.1软件,得到共同靶点,即四妙丸作用类风湿关节炎的靶点;通过Cytoscape 3.7.0软件构建成分-靶点网络,并利用软件中Network Analyzer插件分析网络节点度和介数中心度;最后,通过DAVID在线分析工具对靶标进行基因本体(GO)功能富集分析和生物途径(KEGG)通路富集分析。结果:四妙丸共收集52个有效化合物,并作用于14个类风湿关节炎相关的靶点。成分-靶点网络图显示前列腺素G/H合成酶2、转录因子AP-1及肿瘤坏死因子是四妙丸治疗RA的关键靶点,GO功能富集分析显示126个生物过程、8个细胞组成、5个分子功能,KEGG通路富集分析显示35条通路。结论:四妙丸通过一氧化氮生物合成过程的正调节、钙调蛋白1-单加氧酶活性的正调节、发热的正调节、炎症反应等生物过程,并涉及细胞因子活性、生长因子活性、酶结合等分子功能发挥治疗类风湿关节炎的作用,且肿瘤坏死因子信号通路的调控是四妙丸治疗类风湿关节炎的关键机制之一。Objective:Based on the network pharmacological method,to explore the mechanism of Simiao Wan(四妙丸)in the treatment of rheumatoid arthritis.Methods:The chemical constituents of Simiao Wan were screened and the targets were predicted by TCMSP database with oral bioavailability and pharmacokinetics as the limiting conditions.The targets related to rheumatoid arthritis were retrieved by GeneCards database,and the predicted targets of Simiao Wan and targets related to rheumatoid arthritis were put into the VENNY 2.1 software to get the common targets,namely the targets of Simiao Wan for rheumatoid arthritis.The component-target network was constructed by Cytoscape 3.7.0 software,to analyze the network node degree and the median centrality by using Network Analyzer plug-in software.Finally,functional enrichment analysis of gene ontology and KEGG pathway enrichment analysis were made by DAVID.Results:A total of 52 effective compounds were collected from Simiao Wan and acted on 14 targets related to rheumatoid arthritis.Component-target network showed that prostaglandin G/H synthase 2,transcription factor AP-1 and tumor necrosis factor were the key targets of Simiao Wan in the treatment of rheumatoid arthritis.GO functional enrichment analysis showed 126 biological processes,8 cell components and 5 molecular functions.KEGG pathway enrichment analysis showed 35 pathways.Conclusion:Simiao Wan can play a role in the treatment of rheumatoid arthritis through the positive regulation of nitric oxide biosynthesis,calmodulin-1-monooxygenase activity,positive regulation of fever,inflammatory reaction and other biological processes,involving the molecular functions of cytokine activity,growth factor activity,enzyme binding,and so on.The regulation of signal pathway is one of the key mechanisms of Simiao Wan in the treatment of rheumatoid arthritis.
