摘  要:白色念珠菌是条件致病菌,形成生物膜后耐药性增加,从放线菌次级代谢产物中获得能抑制生物膜形成的蛋白有一定应用前景。采用硫酸铵盐析法提取发酵液中的粗蛋白;微孔板半定量法检测菌株TRM43355发酵液粗提物、粗蛋白及不同理化因素处理后的粗蛋白对白色念珠菌生物膜形成能力的影响。结果显示,菌株TRM43355的粗蛋白对白色念珠菌生物膜形成具有很好的抑制效果,蛋白浓度为2mg/mL时抑制白色念珠菌生物膜形成效果最好,其抑制率可达92%以上。蛋白耐酸,但不耐高温,80℃以上的高温蛋白失活。经有机溶剂和金属离子处理后的粗蛋白对白色念珠菌生物膜的抑制率都有所提高。菌株TRM43355发酵液中的蛋白具有较强抑制生物膜活性。Candida albicans is a conditioned pathogen with increased drug resistance after the formation of biofilms. It has certain application prospects to obtain proteins from actinomycetes secondary metabolites that can inhibit the formation of biofilms. The protein in fermentation broth of TRM 43355 was extracted by ammonium sulfate salting method. Effects of fermentation broth extract of strain TRM43355, crude proteins and crude proteins treated by different physical and chemical factors on the biofilm formation of C. albicans were detected by microporous plate semi-quantitative method. The crude protein of strain TRM 43355 has a good inhibition effect on the formation of C. albicans biofilm, and the best inhibition effect concentration was 2mg/mL, with the inhibition rate of more than 92%. The protein is acid resistant, but dones not have high temperature resistance. The protein in the high temperature environment at 80℃ or above is inactivated.The inhibition rate of crude protein treated with organic solvent and metal ion to candida albicans biofilm was improved. The protein in the fermentation broth of strain TRM43355 had a strong inhibitory effect on C. albicans biofilm formation activity.
