摘  要:肝脂肪变性是甘油三酯以脂滴的形式大量沉积于肝细胞内为特征的一种慢性肝病,是系列严重进展性肝病的最初表现形式,若不加以干预可由单纯的脂肪变性发展为肝炎、肝纤维化、肝硬化,甚至肝癌。目前抗脂肪肝药物研发相对滞后,构建合适的肝脂肪变性体外实验模型对于研究肝病发病机制及快速筛选候选药物具有重要意义。体外肝脂肪变性细胞模型具有实验条件相对稳定、周期短、重复性好、影响因素少等优势,为抗脂肪肝药物筛选和评价提供重要技术手段。本文主要综述了由高能物质、药物、化学物质及病毒这几种常见病因所诱导的肝脂肪变性细胞模型及其致病机制,为研究肝脂肪变性及相关药物的干预作用提供体外实验模型选择参考。Hepatic steatosis is a liver disease characterized by excessive accumulation of triglycerides in hepatocytes in the form of lipid droplets. The hepatic steatosis is an initial manifestation of severely progressive liver diseases and may develop from simple steatosis into the steatohepatitis,liver fibrosis,cirrhosis and even liver cancer if the hepatic steatosis is not intervened efficaciously at the early stage. However,due to the lack of proper models,the development of therapeutic drugs is limited. Therefore,the suitable experimental models in vitro are significant for the pathogenesis study of liver diseases and the fast screening of related drugs. Cellular models in vitro possess the advantages in keeping the stable experimental conditions,short running times,good repeatabilities and fewer influencing factors,which provides an important technical means for the screening and evaluation of anti-fatty liver drugs. In this paper,we mainly review the characteristics and pathogenesis of cellular steatosis models induced by the energetic substances,drugs,chemicals and viruses,so as to provide a reference for the selection of cellular models to investigate pathogenesis of steatosis and evaluate anti-fatty liver drugs.
