摘　　要：目的探讨在抗病毒治疗的HIV/AIDS患者外周血单个核细胞中叉头状转录因子P3(Foxp3) mRNA相对表达量和患者疾病进展的关系。方法采集丽水辖区内95例正接受高效抗病毒(HAART)治疗的HIV/AIDS患者及83例健康对照者的抗凝全血,流式细胞技术检测CD4细胞的数量,荧光定量PCR技术检测HIV核酸、Foxp3 mRNA表达量,对Foxp3 mRNA表达量高于检测下限的患者进行分析。结果在接受抗病毒治疗的95例患者中,患者病毒载量高于检测下限的仅5例。Foxp3 mRNA高于检测下限的有60例。在60例患者中,Foxp3 mRNA基因的表达水平与CD4细胞数量呈正相关。结论对于接受抗病毒治疗的人群,通过检测外周血Foxp3 mRNA的相对表达水平可以推测机体在疾病变化中的免疫状态,Foxp3 mRNA基因的表达水平可以作为预测疾病严重程度的参考指标之一,为患者预后评估及临床治疗开辟了新思路。Objective To investigate the relationship between the relative expression of forkhead transcription factor 3( P3)mRNA in peripheral blood mononuclear cells and disease progression in HIV/AIDS patients with antiviral treatment.Methods The whole blood was collected from 95 HIV-infected patients undergoing HAART treatment and 83 healthy controls in Lishui. The number of CD4 cells was measured by flow cytometry,and fluorescent nucleic acid PCR was used to detect the expressions of HIV nucleic acid and Foxp3 mRNA. The cases in which Foxp3 mRNA was above the detection limit were conducted for analysis. Results Of the 95 patients who received antiviral therapy,only 5 cases' viral load was above the lower limit of detection. Foxp3 mRNA was higher than the detection limit in 60 patients,in which Foxp3 mRNA expression was positively correlated with the number of CD4 cells. Conclusion For people receiving antiviral therapy,the immune status of the body can be predicted by detecting the relative expression level of Foxp3 mRNA in peripheral blood. The expression level of Foxp3 mRNA gene can be used as one of the reference indicators for predicting the severity of disease,which has opened new ideas for patients' prognosis and clinical treatment.
