摘 要:目的:研究白花蛇舌草注射液对人卵巢癌细胞HO-8910PM生物学行为的影响。方法:利用MTT法研究不同浓度白花蛇舌草注射液对人卵巢癌细胞HO-8910PM的生长抑制作用;应用Transwell小室实验检测肿瘤细胞的侵袭能力和迁移能力,流式细胞术分析白花蛇舌草注射液对人卵巢癌细胞HO-8910PM凋亡情况;利用Western blot检测B淋巴细胞瘤-2基因(Bcl-2)、细胞程序性死亡的蛋白质(Bad)、活化的含半胱氨酸的天冬氨酸蛋白水解酶(cleaved caspase-3)、活化的DNA修复酶(cleaved-PARP)细胞蛋白表达变化。结果:与对照组相比,白花蛇舌草注射液各浓度组能显著抑制HO-8910PM细胞增殖率、侵袭能力和运动能力(P<0.05或P<0.01),且随着药物浓度的增加和作用时间的延长,其抑制作用增强;白花蛇舌草注射液12.5,25,50,100 ml·L^-1浓度组的凋亡细胞比例显著增加(P<0.01);白花蛇舌草注射液各浓度组细胞抑凋亡蛋白Bcl-2表达量显著降低,促凋亡蛋白Bad及下游凋亡信号传递和执行细胞凋亡的cleaved caspase-3蛋白和PARP蛋白显著升高(P<0.01),作用效果与药物浓度呈正相关。结论:白花蛇舌草注射液能够抑制人卵巢癌细胞HO-8910PM的侵袭和迁移能力,通过诱导细胞凋亡来抑制细胞增殖。Objective: To study the effects of Hedyotis diffusa injection on the malignant biological behaviors of human ovarian cancer cell line HO-8910PM.Methods: MTT assay was used to study the growth inhibitory effects of different concentrations of Hedyotis diffusa injection in human ovarian cancer cell line HO-8910PM.Transwell chamber assay was used to detect the invasion and migration abilities of tumor cells.Flow cytometry was used to analyze the apoptosis of HO-8910PM cells affected by different concentrations of Hedyotis diffusa injection.Western blot assay was used to determine the changes of cellular protein expressions of Bcl-2,Bad, cleaved-caspase-3 and cleaved-PARP.Results: Compared with the control group,Hedyotis diffusa injection at each concentration could inhibit the proliferation rate,invasion ability and motor ability of human ovarian cancer cell line HO-8910PM (P<0.05 or P<0.01).With the increase of drug concentration and the prolongation of action time,the inhibitory effects were enhanced.The proportions of apoptotic cells in the concentration groups of 12.5,25,50 and 100 ml·L^-1 Hedyotis diffusa injection were significantly increased (P<0.01).The expression of antiapoptotic protein Bcl-2 was significantly decreased in all concentrations of Hedyotis diffusa injection while the expressions of proapoptotic protein Bad and downstream apoptotic signaling cleaved caspase-3 and PARP proteins were significantly increased (P<0.01),and the effects were positively correlated with the drug concentration.Conclusion: Hedyotis diffusa injection can inhibit the invasion and migration of human ovarian cancer cell line HO-8910PM and inhibit cell proliferation by inducing apoptosis.
