高效液相色谱法测定日用香精中的10种香豆素及其衍生物含量

摘　　要：建立了高效液相色谱法同时测定日用香精中10种香豆素及其衍生物含量的分析方法。样品经碱性甲醇超声提取,用Phenomenex Luna C18柱分离,以乙腈和0.1%NH 4Ac溶液为流动相进行梯度洗脱,采用二极管阵列检测器同时在280 nm、306 nm和320 nm的波长下检测。结果表明:香豆素及其衍生物在1.0~100.0μg/mL质量浓度范围内,线性关系良好,相关系数(r^2)在0.9997~0.9999之间;检出限为0.15~0.90 mg/kg,定量限为0.45~3.0 mg/kg;加标回收率和相对标准偏差分别为90.1%~98.3%和1.1%~4.6%。本方法操作简单、回收率高、精密度好,适用于日用香精中禁用香豆素及其衍生物的日常检测。A high performance liquid chromatography based method was established for the determination of 10 coumarin and their derivatives in fragrance compound.The samples were ultrasonic extracted with alkaline methanol.The extract was separated on Phenomenex Luna C 18 column with gradient elution by using acetonitrile and 0.1%ammonium acetate solution as eluant,and then monitored under the detection wavelength of 280 nm,306 nm,and 320 nm by diode array detection method.The results showed that the linearity of 10 coumarin and their derivatives ranged from 1.0μg/mL to 100.0μg/mL with correlation coefficients(r^2)of 0.9997-0.9999.The limit of detection was 0.15 to 0.90 mg/kg,and the limit of quantification was 0.45-3.0 mg/kg.The recoveries of these coumarin ranged from 90.1%to 98.3%,while the relative standard deviations ranged from 1.1%to 4.6%.This method is simple with high recovery and good precision,and suitable for routine determination of coumarin and their derivatives in fragrance compound.
