摘 要:为观察川陈皮素对糖尿病肾病的治疗作用,本研究选用120只SD大鼠适应性喂养2周后分为正常组(20只)和糖尿病肾病造模组(100只),成功构建糖尿病肾病模型后选取50只,分为模型组,川陈皮素低剂量、中剂量和高剂量组以及阳性药物贝那普利组,每组10只,治疗6周后处死,收集尿液检测24h-尿量和24h-尿蛋白,收集血液检测血糖、胰岛素、血脂、肾功能指标和炎性因子的变化特点,收集肾脏检测肾脏病理学以及肾脏组织中凋亡相关蛋白Bcl-2、Bax、Caspase-3表达水平。结果显示,模型组有明显肾小球增大、部分系膜增生和间质纤维化,相较于模型组,贝那普利组和川陈皮素三个剂量组肾小球病变减轻;与模型组相比,贝那普利组和川陈皮素低、中、高剂量组UCr、24h蛋白尿、BUN、Scr、血糖、TG、TC、IL-1、IL-6和TNF-α明显降低(P<0.05),胰岛素含量明显升高(P<0.05);与模型组相比,贝那普利组和川陈皮素低、中和高剂量组Bax和Caspase-3明显降低(P<0.05),Bcl-2明显升高(P<0.05)。上述研究表明,川陈皮素对糖尿病肾脏损害大鼠肾功能具有明显的保护作用。Observe the therapeutic effect of Nobiletin on rats with diabetic nephropathy.120 SD rats were divided into the normal group (20 rats) and diabetic nephropathy model group (100).After the model was successfully constructed,50 rats were selected and divided into the model group,low-dose,medium-dose and high-dose Nobiletin groups and the benazepril group( n =10).After treatment for 6 weeks, they were sacrificed,their urine was collected to detect the 24 h urine volume and24 h-urine protein,and their blood to observe the blood glucose,insulin,blood lipids,renal function indicators and inflammation indexs,at last the renal pathology and the expression of apoptosis-related proteins Bcl-2,Bax and Caspase-3 in kidney tissues were detected.The results of HE staining could be seen the glomerular enlargement,partial mesangial hyperplasia and interstitial fibrosis could be observed in the model group.Compared with the model group,the slices in the benazepril group and nobiletin group were improved.Compared with the model group,the benapril group and the nobiletin low,medium and high dose group for the UCr,24h proteinuria,BUN,Scr,blood glucose,TG,TC,IL-1,IL-6 and TNF-α were significantly decreased ( P <0.05),and insulin was significantly increased ( P <0.05).Compared with the model group,in the benazepril group and nobiletin group the Bax and Caspase-3 were significantly decreased ( P <0.05) and Bcl-2 was significantly increased ( P <0.05).Above results suggested the nobiletin has obvious protective effects on renal function in rats with diabetic kidney damage.
