接受自体动静脉内瘘术的兔动静脉吻合口静脉端颈外静脉内膜厚度及自噬相关蛋白表达观察  

Intimal thickness and autophagy-related protein expression in external jugular vein of arteriovenous anastomosis in rabbit undergoing autogenous arteriovenous fistula

摘　　要：目的 观察接受自体动静脉内瘘(AVF)术的兔动静脉吻合口静脉端颈外静脉内膜厚度及自噬相关蛋白Beclin-1、LC3-Ⅱ、P62的表达情况。方法 30只雄性新西兰兔,麻醉后切开皮肤,解剖分离出左侧颈总动脉和颈外静脉,进行端-侧吻合,记为实验组;同时解剖分离右侧颈总动脉和颈外静脉,不予处理,记为对照组。分别于术后第1周(T1)、第2周(T2)、第4周(T3)时,实验组在吻合口静脉端取一段颈外静脉,对照组在相对应位置取一段颈外静脉,采用HE染色法测算兔颈外静脉内膜厚度,采用Westernblotting法检测兔颈外静脉Beclin-1、LC3-Ⅱ、P62蛋白(以荧光强度表示目的蛋白的相对表达量)。结果 实验组T1、T2、T3时兔颈外静脉内膜厚度分别为(28.94±2.36)、(36.71±3.64)、(67.51±6.40)μm,对照组T1、T2、T3时分别为(16.96±3.76)、(17.20±3.68)、(17.12±3.59)μm,两组同一时间点相比,P均<0.05。与对照组同一时间点相比,实验组Beclin-1、LC3-Ⅱ蛋白的相对表达量均升高(P均<0.05),P62蛋白的相对表达量均下降(P均<0.05)。实验组内各时间点相比,Beclin-1、LC3-Ⅱ蛋白的相对表达量在T2、T3时均上升(P均<0.05),P62蛋白的相对表达量在T2、T3时均下降(P均<0.05)。实验组兔颈外静脉内膜厚度与Beclin-1、LC3-Ⅱ蛋白表达均呈正相关(r分别为0.887、0.919,P均<0.01),与P62蛋白呈负相关(r=-0.788,P<0.01)。结论 接受AVF术的兔动静脉吻合口静脉端颈外静脉出现了内膜增生与自噬功能增强,且内膜增生与自噬功能增强呈正相关。Objective To observe the intimal thickness of the external jugular vein and the expression of autophagy-related proteins Beclin-1, LC3-Ⅱand P62 in rabbits undergoing autogenous arteriovenous fistula (AVF). Methods Thirty male New Zealand rabbits were anesthetized and the skin was dissected. The left common carotid artery and the external jugular vein were dissected and the end-to-side anastomosis was performed, which was taken as the experimental group. The right common carotid artery and external jugular vein were dissected, which was taken as the control group. At the first week (T 1), the second week (T 2), and the fourth week (T 3), we took a segment of the external jugular vein at the end of the anastomotic vein in the experimental group, and took a segment of the external jugular vein at the corresponding position in the control group. The intimal thickness of rabbit external jugular vein was measured by HE staining. Beclin-1, LC3-Ⅱ, and P62 proteins were detected by Western blotting (the relative expression of target protein by fluorescence intensity). Results The intimal thickness of the external jugular vein at T 1, T 2 and T 3 was (28.94±2.36),(36.71± 3.64 ) and (67.51±6.40)μm, respectively, in the experimental group, versus (16.96±3.76),(17.20±3.68), and ( 17.12 ±3.59)μm, respectively, in the control group, with statistically significant difference ( P <0.05). Compared with the control group, the relative expression of Beclin-1 and LC3-Ⅱprotein in the experimental group increased (both P <0.05), and the relative expression of P62 protein decreased ( P <0.05). In the experimental group, the relative expression levels of Beclin-1 and LC3-Ⅱincreased at T 2 and T 3 (both P <0.05), and the relative expression of P62 decreased at T 2 and T 3 (both P <0.05). The intimal thickness of rabbit external jugular vein was positively correlated with the expression of Beclin-1 and LC3-Ⅱ( r =0.887, 0.919, both P <0.01), and was negatively correlated with P62 protein ( r =-0.788 , P <0.01).
