益气活血化瘀方联合康脉胶囊对下肢缺血再灌注损伤大鼠炎性反应与氧化应激及细胞凋亡影响

摘　　要：目的:分析益气活血化瘀法联合康脉胶囊对下肢缺血再灌注损伤大鼠炎性反应与氧化应激及细胞凋亡影响。方法:选取SD雄性大鼠60只,随机数字表法将大鼠分成假手术组、模型组、阳性对照组和联合用药组,每组各15只,除假手术组外,其他各组大鼠制备下肢缺血再灌注损伤模型,术后阳性对照组腹腔注射剂量3mg/kg依达拉奉,联合用药组灌胃剂量3mg/kg康脉胶囊药液和2ml益气活血化瘀方药液,假手术组及模型组大鼠分别灌胃2ml生理盐水,每天1次,连续灌胃7天。ELISA法检测血清白细胞介素-1(IL-1)、IL-6及肿瘤坏死因子-α(TNF-α)、谷胱甘肽过氧化物酶(GSH-PX)、丙二醛(MDA)、髓过氧化物酶(MPO)、超氧化物歧化酶(SOD)含量,RT-PCR检测大鼠左胫前肌bax、bcl-2mRTA相对表达量,并观察大鼠骨骼肌细胞病理状况。结果:和假手术组相比,模型组大鼠血清TNF-α、IL-6及IL-1含量上升;和模型组相比,阳性对照组、联合用药组大鼠血清TNF-α、IL-6及IL-1含量下降,差异均有统计学意义(P<0.05);和假手术组相比,模型组大鼠血清GSH-PX、SOD含量下降,MPO、MDA含量上升;和模型组相比,阳性对照组、联合用药组大鼠血清GSH-PX、SOD含量上升,MPO、MDA含量下降,差异均有统计学意义(P<0.05);和假手术组相比,模型组大鼠bax mRNA表达上升,bcl-2mRNA表达下降;和模型组相比,阳性对照组、联合用药组大鼠bax mRNA表达下降,bcl-2mRNA表达上升,差异均有统计学意义(P<0.05)。结论:益气活血化瘀方联合康脉胶囊可有效降低下肢缺血再灌注损伤大鼠机体内氧化应激与炎性反应程度,抑制细胞凋亡,降低机体受损程度。Objective:To analyze the effects of Yiqi Huoxue Huayu combined with Kangmai Capsule on inflammatory response,oxidative stress and apoptosis for rats with ischemia-reperfusion injury of lower limbs.Methods:Sixty SD male rats were randomly divided into sham operation group,model group,positive controlled group and combination group,with 15 rats in each group.The rats in the other groups were prepared for the lower limbs except the sham operation group.The Ischemia-reperfusion injury model and the postoperative positive controlled group,intraperitoneal injection of 3 mg/kg edaravone,combined the medication group,intragastric dose of 3 mg/kg Kangmai Capsule Liquid and 2 ml Yiqi Huoxue Huayu Recipe,sham operation rats in the group and the model group were intragastrically administered with 2 ml of normal saline once a day for 7 days.Detection of serum interleukin-1(IL-1),IL-6 and tumor necrosis factor-α(TNF-α),glutathione peroxidase(GSH-PX),malondialdehyde(MDA)by ELISA.The contents of bax and bcl-2 mRTA in the left anterior tibialis anterior muscle were detected by RT-PCR.The pathological changes of rat skeletal muscle cells were observed.Results:Compared with the sham operation group,the serum levels of TNF-α,IL-6 and IL-1 in the model group increased.Compared with the model group,the serum of the controlled group and the combination group were serum TNF-α,IL-6 and The IL-1 content decreased,the difference was statistically significant(P<0.05).Compared with the sham operation group,the serum GSH-PX and SOD levels in the model group decreased,and the MPO and MDA contents increased.Compared with the model group,The levels of serum GSH-PX and SOD in the positive controlled group and the combination group increased,and the contents of MPO and MDA decreased,the difference was statistically significant(P<0.05).Compared with the sham operation group,the expression of bax mRNA in the model group was higher than that in the sham operation group.The expression of bcl-2 mRNA decreased.Compared with the model group,the expre
