健脾养肝方对大鼠肝癌模型肝功能及微环境的影响

摘　　要：目的:分析健脾养肝方对大鼠肝癌模型肝功能及微环境的影响。方法:选用健康SPF级雄性Wistar大鼠60只,将其随机分为阴性对照组、模型组、大剂量健脾养肝方组,小剂量健脾养肝方组,每组15只,除对照组外三组均以二乙基亚硝胺诱导肝癌病变,健脾养肝方组分别给予大小剂量不同进行干预,连续16周。观察并记录大鼠一般情况及体重变化、存活率及肝癌率、苏木精-伊红染色法(HE法)检验组织病理变化、全自动生化检查肝功能、酶联免疫吸附法(ELISA)测定炎性因子水平。结果:健脾养肝方各组大鼠体重明显高于模型组(P<0.05);大小剂量两组大鼠体重组间无明显差异(P>0.05),健脾养肝方各组存活率均显著高于模型组(P<0.05),大小剂量两组存活率组间无明显差异(P>0.05);健脾养肝方各组肝癌率显著低于模型组(P<0.05),大小剂量两组肝癌率组间无明显差异(P>0.05),与对照组比较其余三组肝功能ALT、AST、GGT、ALP、TBIL均明显升高(P<0.05);健脾养肝方各组ALT、AST、GGT、ALP、TBIL均显著低于模型组(P<0.05),大剂量组肝功能各项指标明显低于小剂量组(P<0.05),与对照组比较其余三组肝功能IL-6L、TNF-α炎性因子均明显升高(P<0.05);健脾养肝方各组IL-6L、TNF-α均显著低于模型组(P<0.05),大剂量组炎性因子各项指标明显低于小剂量组(P<0.05)。结论:健脾养肝方可通过改善肝功能及微环境来延缓或阻断肝癌病变。Objective:To investigate the effect of Jianpi Yanggan Decoction on liver function and microenvironment for liver cancer model rat.Methods:60 healthy SPF male Wistar rats were selected and randomly divided into controlled group,model group,high dose Jianpi Yanggan Decoction group and low dose Jianpi Yanggan Decoction group,each with 15 cases.Rats were made into liver cancer models except those in the controlled group and received the corresponding treatments.16 weeks later,general condition,weight change,survival rate and liver cancer rate,histopathological changes observed by hematoxylin-eosin staining method(HE method),liver function determined by automatic biochemical examination,and inflammatory factor levels detected by enzyme-linked immunosorbent assay(ELISA)were recorded and compared between groups.Results:The body weight of Jianpi Yanggan Decoction group was significantly higher than that in the model group(P<0.05),while no difference was found between the high dose group and low dose group(P>0.05).The survival rate of Jianpi Yanggan Decoction group was significantly higher than that in the model group(P<0.05),while no difference was found between the high dose group and low dose group(P>0.05).The liver cancer rate of Jianpi Yanggan Decoction group was significantly lower than that in the model group(P<0.05),while no difference was found between the high dose group and low dose group(P>0.05).The levels of liver function related indexes including ALT,AST,GGT,ALP and TBIL ranking in an ascending order was the controlled group,the high dose group,the low dose group and the model group,with statistic difference(P<0.05).IL-6 and TNF-αlevels ranking in an ascending order was the controlled group,the high dose group,the low dose group and the model group,with statistic difference(P<0.05).Conclusion:The application of Jianpi Yanggan Decoction can effectively ameliorate the liver function and microenvironment,thus to delay or block liver cancer lesions.
